The research of design,synthesis and properties of triazole compounds has been important in organic synthetic chemistry.The triazole-based carboxylic acid compounds with common characteristics of both triazole and acid can coordinate to metal ions and form various hydrogen bonds as well as have a wide range of biological activities.Therefore,they are widely used in pesticide,medicine,chemical industry,supramolecular materials and molecular recognition.In chapter 1,the research progress in the synthesis,poroperties and application of triazole compounds and their derivatives.Then the synthesis methods of trimethoprim?TMP?are introduced.In chapter 2,fifteen 1,2,3-triazole carboxylic acids and their derivatives were synthesized by diazotization,azide,Claisen condensation,esterification,hydrazine hydrolysis and cyclization reaction from substituted aniline.And the structures of these1,2,3-triazole derivatives were characterized by IR and 1H NMR.The optimum synthesis conditions of 1,2,3-triazole compounds were studied for the different substited aniline.In chapter 3,the fluorescence properties of the carboxylic ester compound?i?were studied in the ethanol/water binary solution.The results show that the compound?I?can recognize Fe3+and Cr2O72-,and the recognition effect is not interfered by other ions.The detection limit of?i?to the Fe3+and Cr2O72-is 56.6 uM?18.86 ppm?and 8.33uM?1.8 ppm?,respectively.In Chapter 4,synthesis,characterization and electrochemical properties of the copper complex,which was obtained by a triazole carboxylic acid compound and the copper nitrate,were studied.The complex has the strongest electrochemical signals in the AcOH-AcONA buffer solution with a pH of 5.5.The electrochemical study on the interaction between the copper complex and the herring sperm DNA indicates that the electrochemical signal of the complex can be reduced after the herring sperm DNA was added,and the reaction between the complex and the dsDNA and ssDNA is mainly controlled by the diffusion precess.The binding constant of the complex and the herring sperm DNA was 2.577×103 L·mol-1.In chapter 5,the technological improvement of trimethoprim is studied,and the optimum technological conditions are obtained based on the analysis of many experimental data.In the first step,the optimum ratio of the isomers was,raw material:sodium methoxide:ethylene glycol monomethyl ether=1:0.5:1,and the experimental conditions were 80-85?,refluxing for 3 h;In the second step of cyclization reaction,the optimum ratio was,raw material:sodium methoxide:guanidine nitrate=1:2:2,the experimental condition were 90-100?,refluxing for 7h.
