| In this study,water chestnuts were selected as the research object,through the selection of preservatives and heat treatment conditions to determine the impact of its cold storage quality.And taking water chestnut peel extract as the research object,through in vitro and in vivo antioxidant tests,and compared with antioxidants,the in vitro antioxidant activity and antioxidant effect in vivo were investigated.The main contents and conclusions are as follows:In the first part,through the use of preservatives and heat treatment processing fresh cut chufa,determination of chufa pulp color,decay rate,change rate,soluble solids,soluble protein content and ascorbic acid content of weightlessness,analysis of different concentrations of preservatives and heat treatment of a combined effect on the storage quality of fresh cut Chinese water chestnut.After comparison test,1.5% sodium diacetate was chosen as the preservative of fresh-cut water chestnuts.Fresh-cut water chestnuts were treated with the same heat treatment at 55 ℃ and 65 ℃ for 20 min.The results showed that in the experiment of chroma and soluble protein,the effect of 55 degree heat treatment group was better.In the test of decay rate,weight loss rate,soluble solids and VC,the effect of 65 degree heat treatment group was better.In the second part,the extract of Eleocharis tuberosa peel extract(ETPE)as material was investigatedeffects of reduction ability and its ability to scavenge DPPH-,O2--,OH three free radicals,and ascorbic acid(VC)and 2,6-tert butyl-4-methyl phenol(BHT),which was also studied capacity of antioxidant in lard..The reducing power and DPPH· free radical scavenging power of water chestnuts peel extract were weaker than that of VC and BHT,but the extract of water chestnut peel was 100mg/L~140mg/L,which was equivalent to that of VC.The scavenging ability of extract from water chestnut peel to O2-radical was similar to that of BHT.Its scavenging ability to OH radical was stronger than that of VC,and its antioxidant ability in lard was lower than BHT,but stronger than VC.In the third part,we used three different dose levels(High dose of 200mg/kg BW d,medium dose of 100mg/kg BW d,low dose of 50mg/kg BW d)of the extract of Eleocharis tuberosus,Total superoxide dismutase(T-SOD),glutathione peroxidase(GSH-PX),and hydrogen peroxideEnzyme(CAT)in the serum and liver tissues were determined by intragastric administration of the positive control(vitamin E)activity,total antioxidant capacity(T-AOC)and malondialdehyde(MDA)content in mice were measured to explore its anti-oxidative effects in vivo.The activity of T-SOD in the serum,liver,kidney,high-dose group of antioxidant effect is better in the heart,the effect of the middle dose group is better;The activity of CAT in the liver,kidney,heart,high dose group is better,in the serum,the effect of the middle dose group is better;T-AOC in the serum,liver,heart,high-dose group is better,in the kidney,the effect of the middle dose group is better;The effect of GSH-PX in serum and kidney in high dose group was better than that in low dose group.The effect of GSH-PX was better in the middle and middle dose groups.The MDA content in the blood,liver,kidney and heart,the water chestnut peel extract high dose group the best,and stronger than the VE positive control group. |
