Ziziphus jujuba Mill.,a plant of the genus Ziziphus Mill in the family of Rhamnaceae.Jujube is a nourishing medicine and its pharmacological activity is extensive.The effect of liver protection and anti-cancer is particularly obvious.For its fruit,leaves,seeds,bark and root can be used for the medicine.According to the literatures,there are many research which has reported on the chemical composition of the jujube fruit pulp.However,little research reported on the chemical compositions of jujube seeds.The survey found that a large number of jujube seeds were treated by the waste landfill or fuel after the jujube processed,which causing the resource waste and environmental pollution.The historical material records that jujube seeds has the curative effective of thirst quenching,cure boils,moistens the intestines and promotes digestion.We studied on the chemical composition of jujube seeds and analysis,expecting to get the chemical composition,which has important bioactivity.It will be of great practical significance to the development of traditional Chinese medicine resources.We collected jujube seeds from Jiaxian in the northern of Shaanxi province as the research object,used silica gel,CHP20P and polyamide of macroporous adsorption resin,Sephadex LH-20 column chromatography technology.We have isolated 20 compounds from the ethanol extract of jujube seeds.According to the physical and chemical properties and we also have used the modern spectrum technology(1H-NMR?13C-NMR?HPLC-UV?ESI-MS,etc.)and HPLC or TLC.Then identified 19 compounds,including 3,3'-di-O-methylellagic acid-4-O-?-D-glucopyranoside?1??5-hydroxymethyl-furoic acid?2??catechin?3??zizybeoside??5??naringenin?6??kaempferol?7???E?-3,3'-dimethoxy-4,4'-dihydroxystilbene?8??isovanillin?9??palmitoleic acid?10??3,3'-di-O-methylellagic acid?11??daucosterol?12??protocatechuate?13??gallic acid?14??dihydrocharcone-4'-?-D-glucopy ranoside?15??3-oxo-oleanoic acid?16??scopoletin?17??4-hydroxy-3-methoxybenzoic acid?18???,?-D-glucopyranoside?19??dehydrodigallic acid?20?.The structure of compound 4 is still under identification,and all compounds were isolated from the jujube seeds from Jiaxian for the first time.In addition,an effective HPLC method was established for the first time to measure the average content of the nine compounds in the ethanol extract of jujube seeds which was isolated from the jujube seeds from jiaxian county.The results of methodological verification show that the method was accurate and reproducible.In addition,compounds with significant inhibitory effects on?-glucosidase,tyrosinase and acetylcholinase were screened out through the enzyme activity inhibition experiment,which is conducive to the further study of its application in the fields of hypoglycemic and anti-alzheimer drugs and cosmetics.
