| Grape(Vitis vinifera L.)is a woody vine plant of Vitis L.(Vitaceae Juss.),which is widely cultivated all over the world.This thesis summarizing the plant profile,chemical composition,bioactivities,and current status of its development and utilization.This research will provide reference for studying the major chemical of grape leaves.Red grape leaves were collected from Weinan,Shaanxi Province.Twelve main compounds from the petroleum ether fraction of red grape leaves were identified by GC-MS.Among them,the relative contents of vitamin E,squalene-3-ol,andα-linolenic acid were 19.39%,17.88%,and 10.78%respectively.The 90%ethanol/aqueous extract was isolated by using normal phase,and reverse phase column chromatography technology.The characterization of chemicals was developed by TLC,HPLC,UV,13C-NMR,and 1H-NMR.Totally,24compounds were isolated and characterized,including isorhamnetin-3-O-glucuronide(1),rutin(2),(3R,4S)-3,4,8-trihydroxy-3-methyltetralone(3),panose(4),naringin(5),astragalin(6),isoquercitrin(7),quercetin-3-O-β-D-glucopyranoside(8),kaempferol-3-O-α-L-rhamnoside(9),isorinic acid methyl ester(10),4,5-dicaffeoylquinic acid(11),dimethyl malate(12),caiophoraenin(13),quercetin(14),kaempferol(15),methyl-6-methylheptanoate(16),α-linolenic acid(17),methyl elaidate(18),3-α-isomaltosyl-α,α-trehalose(19),25-deoxyecdysone-3-O-D-glucopyranoside(20),dihydrovomifoliol-9-O-β-D-glucopyranoside(21),astilbin(22),quercetin-3-O-β-D-glucuronide-6’’-methyl ester(23),and quercetin-3-O-[2’’’,6’’’-O-diacetyl-β-D-glucopyranosyl-(1→6)-β-D-glucopyranoside](24).Compounds 3,4,5,7,10,11,12,13,16,19,20,21,22,23,24 were isolated from red grape leaves for the first time.A quantitative analysis method by using high performance liquid chromatography of 10 main compounds in red grape leaves was first established and verified.The content of(3R,4S)-3,4,8-trihydroxy-3-methyltetralone is the highest,which is 1.02 mg/g.The methodological verification results show that the analytical method has great linearity,precision,repeatability,stability,and recovery rate.Also,the activity inhibition experiments of tyrosinase,acetylcholinesterase,andα-glucosidase were developed to all compounds.Flavonoids,unsaturated fatty acids,and phenolic acids demonstrated effectively tyrosinase activity inhibition.Flavanols performed good inhibitory of the acetylcholinesterase andα-glucosidase activities. |
