As a natural yellow pigment,the pigment of “Laba” garlic has great potential for application.In recent years,the preliminary exploration of the yellow pigment of “Laba”garlic has been carried out,but its formation route and structure are still in the speculative stage.Further study on the structure of the yellow pigment and reveal its stability and cell transport mechanism are of great significance for the processing and utilization of the yellow pigment.This project has made the following progress on the basis of previous research:(1)Using yellow crushed garlic as raw material,the solvent was extracted by solvent extraction method,combined with alcohol precipitation,extraction,column chromatography to remove impurities,and further separated and purified by liquid chromatography.Identification and analysis were carried out using an ultraviolet spectrophotometer and ultra performance liquid chromatography mass spectrometry(UHPLC-MS).The pigment of“Laba” garlic has a characteristic absorption peak at 440 nm,the relative molecular weight is228.(2)Thermogravimetric analysis(TGA),differential scanning calorimetry(DSC)and in vitro digestion simulation revealed the thermal stability and the stability in gastrointestinal digestion of the pigment of “Laba” garlic.There were two weight loss in T?140°C and174??T?244?.A yellow pigment degradation kinetics model was constructed to predict the degradation trend of yellow pigment of “Laba” garlic at different times and temperatures.The yellow pigment was degraded by 14.82% and 35.51% in gastric digestion and intestinal digestion,respectively,and yellow pigment was more unstable in the intestinal environment.(3)Caco-2 cell model was established and the cell model was validated for transport experiments by three morphological observations of cell morphology,transmembrane resistance and sodium fluorescein flux.When the concentration of the yellow pigment was less than 5 mg/m L,there were no toxic effect on Caco-2 cells.Further study on the effects of pigment concentration,transport time and inhibitors on the transport of yellow pigment.At the experimental concentration and time,the cumulative transport capacity of the yellow pigment was proportional to the transport concentration and time.The transport mode was mainly passive transport,and its Papp(AP-BL)was 1.30×10-5cm·s-1.It has good absorption in the body.There may be mediated efflux proteins during transport and were affected by energy inhibitors,but were not mediated by P-glycoproteins and multidrug resistance proteins.
