Due to the free active thiol group on its side chain,tiopronin is widely used in protecting and healing hepatic cells.In recent years,the application of tiopronin has been extended to treatment of skin diseases and anti-rheumatic and reduction side effects of radiotherapy and chemotherapy.With the expansion of tiopronin's application range,it has becoming a more and more important issue to determine tiopronin quickly,accurately and conveniently.Estrogens are a kind of steroid hormones.They are not easy to be degraded in environment.So estrogens existing in the environment of soil and water may be enriched to food chain.Long-term intake of estrogen can cause disorders in both men and women's endocrine systems.It can also affect human's reproductive system and increase the chance of getting cancers.Therefore,establishing a method for simultaneously quantification estrogens in certain food has become a significant research area.Based on these problems,in this paper we studied the analysis method of tiopronin based on fluorescence resonance energy transfer and the analysis method of estrogen residues in honey by ultra performance liquid chromatography-tandem mass spectrometry.The main conclusions are as the following:In the first chapter,a brief introduction of the effect and clinical application of thiopronin and an overview of estrogen and its harmness were given.The research status of tiopronin and estrogen's detection technology at home and abroad were elaborated in detail respectively too.In the second chapter,we use gold nanoparticles(AuNPs)and rhodanmine B(RhoB)to establish a fluorescence resonance energy transfer method.The method can be used for rapidly measurement of tiopronin content.The experimental principle was verified.The effects of the volume of gold nanoparticles,the addition order of tiopronin and the fluorescent reagent as well as the incubation time of the reaction between gold nanoparticles and RhoB on experimental results were analyzed.And the experimental parameters were optimized.It was found that the addition order of fluorescent reagent(RhoB)and tiopronin can have obvious influence on the experiment result.The first step is to add tiopronin and then fluorescent agent(RhoB).The effect is more obvious and the measurement sensitivity is better.In the third chapter,to obtain better efficacy,a method for the rapid measurement of tiopronin content was established based on fluorescence resonance energy transfer using gold nanoparticles(AuNPs)and carbon quantum dots(CQDs).The experimental principle was also verified.The effects of the volume of gold nanoparticles and the incubation time of the reaction between gold nanoparticles and CQDs on experimental results were analyzed as well.And the experimental parameters were optimized.Compared to the method using rhodamine B and gold nanoparticle,this method has wider linear range,better linear,better recovery and precision,lower detection limit,but the selectivity is slightly worse.In the fouth chapter,a method for the simultaneous determination of six estrogens(estrone,estradiol,estradiol,estradiol,estradiol,hexestrol and bisphenol A)in honey by solid phase extraction-ultra high performance liquid chromatography tandem mass spectrometry(SPE-UPLC-MS/MS)was established.To reduce the matrix interference and improve the recovery in the determination of six estrogens in honey,four kinds of eluents were used and the quantity of the four eluents was optimized.As a result,the analysis effect was largely improved.The rate of recovery was 78.5~112.9%,RSD was 0.66~8.86%,the detection limit was 0.04~0.19 mg/kg,and the quantitative limit is 0.15~0.62 mg/kg.In the last chapter,we summarized the above reseach and prospected for further research.
