Sulfonamides are a class of molecules containing sulfonyl and amines with a covalent bond, which show important biological activities and have important applications in the field of biomedicine, such as anti-cancer,sterilization,narcotic analgesia, and inhibition of human carbonic anhydrase activities. In recent years, more and more researchers work on sulfonamide, and a lot of curative drugs have been developed, such as celecoxib for the treatment of rheumatoid arthritis, glibenclamide with acute hypoglycemic effect, and anti-migraine agent sumatriptan. It is found that sulfonamide functional group is a special and important functional group.By introducing different functional groups into sulfonamide compounds, some efficient biomedicines and new functional materials can be developed.4-[3-(3-acetylphenyl)ureido]benzsulfamide is an important carbonic anhydrase inhibitor and inhibits the metastasis of breast cancer cells. This paper contains two parts of the work.In the first part,we synthesized 4-[3-(3-acetylphenyl)ureido] benzsulfamide by the addition reaction of p-aminobenzene sulfonamide and 3-acetyl phenyl isocyanate. The condensation reaction of 4-[3-(3-acetylphenyl)ureido] benzsulfamide with tetrahydropyrane-4-carboxylic acid was studied. The influences of solvents, condensation agents and alkalis on the yields were studied, and the optimum reaction conditions were determined. In the second part,a series of 4-[3-(3-acetyl benzene)benzenesulfonamide derivatives were synthesized by the condensation reaction of 28 kinds of carboxylic acids with 4-[3-(3-acetylphenyl)ureido]benzsulfamide,which laid the foundation for further study of their biological activities.
