The rapid development of nanoscience and nanotechnology plays a great role in the widely application of nanomaterials in the biology and medicine fields.Recently, nanomaterials have shown great potential in drug delivery,bio-imaging and photothermal therapy applications due to their small size, high specific surface area and easy modification.Among various synthetic methods, the template method has attracted wide attention because it can accurately control the size, structure, composition and morphology of nanomaterials. In order to prepare expected multifunctional nanomaterials,we need to design a simple synthetic strategy based on the template method and explore its potential application in cancer theranostics. We report a simple and reproducible method for the synthesis of monodispersed biodegradable mesoporous ZnO nanospheres(mZnO NSs)with tunable sizes assembled from ultrasmall particles by using polyacrylic acid(PAA)as template. The obtained mZnO NSs could be responsively decomposed into small fragments under the weak acid environment to meet the requirement for pH-stimulus drug release and in favor of biodegradation. The designed mZnO NSs as drug delivery systems not only achieved the excellent cancer chemotherapy but also could be rapidly decomposed or metabolized to reduce their toxicity.Furthermore, the synthesis method based on PAA template can be extended to the preparation of other composite nanomaterials, providing a broad perspective for future cancer theranostics. In the following work,we explored a unique lactobionic acid(LA)modified metallic one dimensional nanorod/metal organic framework Janus nanoparticles(1D NR/MOF JNPs),LA-gold NR/zeolitic imidazolate framework-8(LA-AuNR/ZIF-8)JNPs,for computed X-ray tomography(CT)imaging guided liver cancer targeted synergistic chemo-photothermal theranostic.Taking advantages of the 1D nanostructure of AuNR,the polyacrilic acid(PAA)was selectively attached to one side of AuNR for further growth of ZIF-8,and the exposed AuNR was modified with targeting agent LA,thus realizing the drug loading and CT imaging guided liver cancer specific targeted theranostic. In addition, the mice treated with doxorubicin(DOX)loaded LA-AuNR/ZIF-8 JNPs under near infrared(NIR)laser irradiation showed significant tumor inhibition, indicating the effective combination of pH and NIR stimuli response release, cancer specific targeting and synergistic chemo and photothermal therapy.
