Ketogenic diets are a group of diets rich in natural fats,but contain an appropriate proportion of protein and limit carbohydrate combinations.Polysaccharide and ketogenic diet have certain effects on the treatment of tumors,but there are few reports on the combination of polysaccharide and ketogenic diet in the treatment of tumors.This paper intends to combine polysaccharide with ketogenic diet,to explore its anti-tumor effect and enhance the immune activity of tumor body were studied in terms of tumor weight,organ index,immune activity and intestinal flora.The main research contents are as follows:1.Four medicinal edible polysaccharides,Lentinus edodes,Astragalus membranaceus,Codonopsis pilosula and Tremella fuciformis were obtained by hot water extraction and ethanol precipitation.The extraction rates were 2.60%,0.42%,8.89% and 8.96%respectively.The polysaccharide contents were 44.49%,31.34%,74.29% and 64.17%respectively.2.After inoculating lung cancer cells,mice were fed ketogenic diet(KD),ketogenic diet + polysaccharides,thirty days later,the weight of tumors in each group was compared.The results showed that the tumors in ketogenic diet group and ketogenic diet combined with polysaccharides group decreased significantly,which was significantly different from those in SD group(P < 0.05).The results showed that ketogenic ketone diet had a certain preventive effect on the growth of tumors;Compared with ketogenic diet group,ketogenic diet combined with four medicines and edible polysaccharides had no significant effect on tumor size(P > 0.05).The body weight of mice fed with ketogenic diet for 30 days showed a decreasing trend,which was significantly different from SD group(P < 0.05);however,there was no difference in body weight between KD group,KD + polysaccharide group.It was found that ketogenic diet could reduce the body weight of mice,the color of hair was not smooth,and the metabolism was slower than normal diet group.3.After 15 days of inoculation of lung cancer cells,mice were fed ketogenic diet and ketogenic diet + polysaccharide,fifteen days later,the weight of tumors in each group was compared.The results showed that the tumors in each group fed ketogenic diet decreased significantly,and there was a significant difference between SD group and ketogenic diet(P < 0.05),indicating that ketogenic diet had a certain therapeutic effect on tumor growth.It was found that the combination of polysaccharide and ketogenic diet and ketogenic diet alone could reduce the weight of mice.There was a significant difference between SD group and SD group(P < 0.05),and the hair color was not smooth,and the metabolism was slower than that of standard diet group.The immunological activity of mice was investigated.It was found that 400 mg/kg of Codonopsis pilosula polysaccharides(DS-400)and 800 mg/kg of Codonopsis pilosula polysaccharides(DS-800)could increase the phagocytic function of macrophages.The secretion of TNF-? in serum of mice was increased by 400 mg/kg of Lentinan(XG-400)and 400 mg/kg of Astragalus membranaceus(HQ-400).4.800 mg/kg of Lentinus edodes polysaccharides combined with ketogenic diet could increase the abundance of intestinal flora in mice.The Shannon index was the highest(5.61).The Shannon index and faith_pd index of DS-400 were the lowest(3.96,10.96),and the alpha diversity index was the best.In intestinal flora,Erysipelotrichi increased in ketogenic diet group,and the highest content of polysaccharide of Codonopsis pilosula 800mg/kg(DS-800)was 29.79%.Lactobacillus was the highest(3.51%)in intestinal flora of DS-800 mice.The combination of Codonopsis pilosula polysaccharide and ketogenic diet increased the number of beneficial flora in intestinal flora of mice,and inhibited the growth of pathogenic bacteria to a certain extent.
