Fe3O4 magnetic nanoparticle?MNPs?have displayed the wide application in the present medicinal field.Especially in cancer therapy,MNPs offer the opportunity of improving the effectivity of the treatment and reduce side effects by magnetic accumulation of MNPs-bound chemotherapeutics in the tumor area.The objective of this work is the synthesis,characterization and property evaluation of magnetic nanovectors coated with?-cyclodextrin??-CD?or its derivatives,and experimental and modelling studies of the efficacy with which such particles can be captured by an external magnetic field.In this work,firstly magnetic Fe3O4 nanoparticles have been covalently attached with?-CD and its derivatives by bifunctional polyethylene glycol to form magnetic nanovectors for delivering 5-fluorouracil?5-FU?,respectively.Then the magnetic microspheres were characterized by infrared spectroscopy?IR?,X-ray diffraction?XRD?,scanning electron microscopy?SEM?.Additionally,their hemolysis and anticancer activities were also examined in the study in vivo and in vitro.In vitro experiments demonstrated that these modified magnetic Fe3O4nanoparticles could be highly versatile drug carriers with sustained release behaviour and sufficient magnetic susceptibility.Moreover,these cyclodextrin-grafted nanoparticles loade with 5-FU showed no hemolytic activity.Rat experiments demonstrated the biological applicability of a panel of nano-sized drug delivery systems as anticancer agents since magnetic drug targeting has been employed to improve localized drug delivery and to enhance drug-therapeutic efficiency in rat tumor.Especially,our observation demonstrated that this systems seems to be very biocompatibles vehicles for the administration of hydrophobic drugs such as 5-FU.In summary,the functionalization of magnetic iron oxide nanoparticles by?-CD or its derivatives to conjugate with bifunctional polyethylene glycol was studied which shows that oligosaccharide-coated carriers have great potential to serve asmagnetic vehicles for drug delivery as well as offers a reference for clinical treatment of cancer.
