Natural macromolecular materials are widely used in biological engineering(including cell and tissue engineering)and immunomodulatory carriers due to their unique immune activity,biocompatibility and excellent metabolic properties.Immunotherapy is one of the most promising methods for tumor therapies,and the process of preparing immune agents(antibodies and cell agents)is inseparable from the theoretical guidance and technical support of chemical engineering.In this paper,natural macromolecular material was used to design and prepare the implantable cell scaffold with immune activation function through chemical modification and processing.Through the signal transmission function of the scaffold material,the maturation of antigen presenting cells was induced,and the anti-tumor activity of cytotoxic T cells was effectively promoted.Three kinds of scaffolds were synthesized by different preparation methods.SEM scanning electron microscope was used to characterize the external morphology and spatial structure of the scaffolds.At the same time,the swelling property of the scaffold was characterized.The results showed that the structure of scaffold material was microporous and the internal space was suitable for cell loading.The swelling test showed that the swelling rate of scaffold was low,it's suitable for implanting.The HeLa-eGFP cell was used to prove that the cell scaffold is excellent in cell loading capacity.MTT method was used to determine the influence of scaffold on dendritic cell(DC)growth,and the results showed that scaffold significantly promoted DC cell proliferation.Polymerase chain reaction(PCR)and Enzyme-linked immunosorbent assay(ELISA)measurement revealed that scaffolds could effectively stimulate DC cells and promote DC cells to transcript and release the cytokines such as TNF-??IL-12?IL-1?,indicating that then the scaffold can increase the antigen presentation process of dendritic cells.In conclusion,we successfully prepared cellular scaffolds with immune stimulation function,providing a strong research basis for the follow-up in vivo experiments.
