Recently,nanoparticles that combine synergistic therapy,real-time imaging and spatiotemporally controlled therapies have attracted substantial attention.In particular,it has been verified that combined photothermal therapy(PTT)with chemotherapy and imaging has become one of the research hotspots in cancer therapy.Herein,we fabricated doxorubicin(DOX)-loaded and ?-cyclodextrin(?-CD)-capped mesoporous platinum nanoparticles(MPNPs)(DOX/MPNPs-AD-CD)for combined chemo-photothermal therapy.DOX/MPNPs-AD-CD has the property of controlled release,which accelerates drug release when stimulated by an acidic tumour microenvironment or near-infrared light.MPNPs also have the characteristics of acid-degradable.On the one hand,the accumulation of MPNPs in the body can be reduced.On the other hand MPNPs released cytotoxic Pt2+,which can damage DNA of cells and enhanced the efficiency of chemotherapy.Furthermore,MPNPs have high photothermal conversion efficiency and photothermal therapy effects.In addition,inherent photoacoustic and photothermal imaging can effectively monitor their distribution in vivo.The experimental results showed that the particle size and pore size of MPNPs are 45 nm and 2 nm,respectively.DOX/MPNPs-AD-CD could enter the cell lysosome,and the released DOX can partially enter the nucleus.The MTT results showed that the cytotoxicity of DOX/MPNPs-AD-CD+NIR was strongest.From the animal experiment results we knew that DOX/MPNPs-AD-CD could be passively targeted to the tumor site.The tumor weight,tumor volume,H&E staining and Tunel experiment showed that the DOX/MPNPs-AD-CD has the strongest anti-tumor effect under the NIR.To sum up,the results of this study provided basic research data for the clinical treatment in cancer.
