The purpose of this study is to provide data reference for the development and application of flavonoids in raspberry leaves.An improved ethanol extraction process was designed by orthogonal experiment and response surface methodology.The preliminary separation of Flavonoids from raspberry leaves and the accurate determination and verification of rutin content were established and completed by HPLC.The optimum extraction process was 66% ethanol,1:12.5 solid-liquid ratio,and reflux at 41.9 ? for 173.3 min.The optimum extraction rate was 1.127%.The optimum extraction rate of response surface methodology was slightly higher than that of orthogonal experiment,and the technological conditions were better than those of orthogonal experiment.The actual content of Rutin in raspberry leaf flavonoids was1.514 mg/g,accounting for 13.4% of the total flavonoid content of crude extracts.It could provide theoretical reference for the later development of raspberry leaf rutin veterinary medicine preparations.This paper mainly takes raspberry leaves as the research object,according to the "Compendium of Materia Medica" records that its stem,leaves and immature fruits can be used as medicine.The further development of by-products such as raspberry leaves was explored and preliminarily studied.The basic structure of flavonoids in raspberry leaves was analyzed,which is of great significance for the development,application and deep processing of active components in raspberry leaves,and also provides guidance for the identification of flavonoids in plants.Raspberry leaves are the by-products of Rubus,a representative medicinal plant of Rubus.Their physiological,biochemical and metabolic functions and biological and pharmacological activities still need to be further explored and discussed.In this paper,the method of isolation and identification of active components of raspberry flavonoids was established by high performance liquid chromatography(HPLC).The extraction and purification of Flavonoids from raspberry leaves werepreliminarily studied,and the optimum extraction and purification process of raspberry flavonoids was determined.By completing the analysis of flavonoids in raspberry leaves by liquid chromatography,the content of Rutin in raspberry leaves was preliminarily determined.Rutin is also a precondition for the chemical synthesis of quercetin in natural flavonoid products.It also has some physiological functions and biomedical pharmacochemical activities.Rutin can be developed as a kind of chemical synthesis drug similar to troxerutin,which will expand its application to a certain direction of veterinary medicine.The rutin established in this experiment is crude flavonoids in raspberry leaves.The analysis method of high performance liquid chromatography in extracts can only expand the collection resources of biomaterials for the later research and development of natural extracts of rutin.The further research on the preparation of rutin for veterinary use of raspberry leaves still needs to be further explored.This will provide a basic method for the development,utilization and quality control of by-products of raspberry and raspberry leaves,as well as flavonoids in natural plants.The study of active ingredients provides basic theoretical guidance.
