Ampelopsis grossedentata is belong to grape family,the snake vitis.The pharmacological active site of A.grossedentata was investigated.A.grossedentata contains a lot of the content of flavonoids and polyphenols,the content of water-soluble polysaccharide and water-soluble protein was relatively low.The water-soluble polysaccharide was the pharmacological active site of medicinal plants.In this paper,the optimum extraction conditions of a polysaccharide from A.grossedentata in Bacong village of Zhangjiajie was studied.The preparation of it's buccal tablet was investigated.The results will be provide basic data for the exploitation and utilization of A.grossedentata.1.The polysaccharide was extracted using hot water and ethanol precipitation method.Single factor experiment and orthogonal test were investigated by taking extraction temperature,extraction time and solid-liquid ratio.The monosaccharide composition of the polysaccharide was analyzed.The results indicated that the optimum conditions were as follows: 95?,1:40 solid-liquid ratio and 1h for extraction.The extraction rate of the polysaccharides was 11.26%.The polysaccharide from A.grossedentata was composed mainly of Man,GlcUA,Glc,Gal and Xyl.Its monosaccharide composition normalized mol % Man/GlcUA/Glc/Gal/Xyl is 29/10/7/18/37.The structure of polysaccharide selenide were characterized by IR.The content of polysaccharide selenide was determined by HPLC.The results showed that it is selenide polysaccharides.The polysaccharide from A.grossedentata showed free radical scavenging activity against DPPH in vitro.2.In order to study extraction ratio of biotransformation of A.grossedentata polysaccharide by fermentation of Bacillus subtilis.The results showed that the metabolic process of biotransformation of A.grossedentata polysaccharide was no stable in conditions of 5% of the inoculation amount and 2%~10% of the solid-liquid ratio.Comparatively speaking,the amount of A.grossedentata polysaccharide extracted was the largest on the 4th day of fermentation.In 2%~8% of the solid-liquid ratio and 5% of the inoculation amount,the extraction ratio of biotransformation of A.grossedentata polysaccharide by fermentation of B.subtilis was the largest.3.The weight loss,disintegration time and hardness of buccal tablet of A.grossedentata polysaccharide were used as indicators,the single factor experiments were used to screen primary and auxiliary raw material,and the orthogonal experiments was used to optimize the optimum preparation processing of A.grossedentata polysaccharide buccal tablet.The results showed that the content of A.grossedentata polysaccharide and the content of mixture of microcrystalline cellulose and maltodextrin have a great influence on tablet hardness.When the content of A.grossedentata polysaccharide was unchanged,with the increase of maltodextrin content in buccal tables,the hardness of buccal tables decreased.When the ratio of microcrystalline cellulose to maltodextrin unchanged,with the increase of content of A.grossedentata polysaccharide in buccal tables,the hardness of buccal tables increased.The results of the orthogonal experiments showed that the optimum formula is: the A.grossedentata polysaccharide content was 20%-30%,the ratio of microcrystalline cellulose to maltodextrin was 1:2-1:3,magnesium stearate content ranged from 0.5% to 0.6%.
