White Cordyceps(Cordyceps militaris Link)is a plant naturally derived from the bedside of the Cordyceps militaris during the breeding of Cordyceps militaris.It has complete plant growth properties and is a new type of plant.It is completely consistent with the growth cycle and growth environment of White Cordyceps.Successful artificial domestication.Through application experiments,it is demontrated that the efficacy of White Cordyceps is similar to that of Cordyceps militaris.However it is easy to breed,low cost,and has high application value.The cultivation and application of White Cordyceps can provide a new way for the development of the medicinal materials industry in Changbai Mountain.The research about it is relatively rare and has high research value.In this paper,ultrasonic-assisted extraction method was used to extract polysaccharides from White Cordyceps.The effects of feed-liquid ratio,extraction temperature,extraction time and ultrasonic power on the extraction rate of polysaccharides from White Cordyceps were studied by single factor experiments.The optimal extraction process is determined by orthogonal experimental design and response surface modeling to optimize the extraction process parameters of the process.The optimal extraction process parameters are extraction time 3 h,extraction temperature 60 °C,ultrasonic power 1600 w,and ratio of material to liquid 1:40.The polysaccharide of White Cordyceps obtained by alcohol precipitation was separated and purified by DEAE-Sepharose and glucose Sephadex G-100 column to prepare pure White Cordyceps polysaccharide.Three distinct main peaks were obtained after washing with water and 0.05 M NaCl,which were named as CKI,CKII and CKIII.The monosaccharide composition of CKII in the polysaccharides of White Cordyceps was determined by PMP-pre-column derivatization high performance liquid chromatography(HPLC).It is determined that there are six mainly kinds of monosaccharides which are Ara(L-arabinose)and Gal(D-galactose),Xyl(D-xylose),Man(D-mannose),Glc(D-glucose)and Rha(L-rhamnose),respectively.Establishing an in vitro anti-inflammatory model with mouse mononuclear macrophages(RAW264.7)and mouse embryonic fibroblasts(NIH3T3)to study the antiinflammatory activity of the extracted polysaccharides of White Cordyceps.To determine whether the polysaccharides of Coprinus comatus have anti-inflammatory activity by detecting the secretion of prostaglandins(PGE2)and NO in RAW264.7 and NIH3T3 celllines without and with powder administration.Using rofecoxib as positive control.The anti-inflammatory effect is dependent on the amount of administration: not administered<1 mg/mL <2 mg/mL <5 mg/mL <50 mg/mL <10 mg/mL < rofecoxib.The optimal dose was 10 mg/mL.In this study the best extraction process for extracting polysaccharides from White Cordyceps by ultrasonic assisted method was explored.The monosaccharide composition of the polysaccharide of White Cordyceps was analyzed.The anti-inflammatory activity of polysaccharides from White Cordyceps was studied.It provides more possibilities for the better application of White Cordyceps.It gives a direction for the study of the efficacy of active ingredients in White Cordyceps and the development of related products.
