Research On Anti-arthritis Products Based On EGCG-GA Protein Nanocomposites

Epigallocatechin gallate?EGCG?is a major anti-inflammatory active ingredient in tea,and has synergistic anti-arthritic effects with glucosamine?GA?.It is expected to develop anti-arthritic foods with excellent effects through nano-technology.?1?In this study,the formulation of EGCG-GA-casein?CS?nanoparticles?EGC-NPs?was prepared and its effect on the level of?1-AGP and the expression of the inflammatory factors TNF-?and IL-1?in serum of CIA-induced arthritis SD rats were determined.The results showed that the anti-arthritic effect of EGC-NPs was similar to that of the positive drug?celecoxib?control group.?2?Food safety of EGC-NPs was evaluated using a three-stage trial?acute oral toxicity,genotoxicity,and 28-day oral toxicity?.Studies have shown that acute oral toxicity of EGC-NPs for rats with a LD₅₀>7.5 g/kg·BW,and for mice LD₅₀>15.0 g/kg·BW,which indicated as actually non-toxicity.In the Ames test,there was no significant difference in the rate of micronucleated cells,and the rate of PCE/RBC between EGC-NPs and negative control groups,and in mouse spermatogenic chromosome aberration test,there ware no significant differences in chromosomal abnormal cell rate,autosomal univalent and chromosomal aberration between EGC-NPs and negative control groups;also EGC-NPs has no mutagenic effect on germ cells of adult mice;which demonstrate EGC-NPs are not genotoxic.For the rats orally administrated EGC-NPs for 28 days,no toxicities were observed since no obvious damage was found based on the index:eyes,body weight,food intake,food utilization rate,hematology,blood biochemistry,urine routine,etc.;and no obvious histopathological changes were observed in anatomical and histopathological observations,which demonstrate that EGC-NPs have no subacute toxicity.Therefore,EGC-NPs are non-toxic as food ingredients based on the above three-stage tests for toxicity.?3?An anti-arthritic food with coconut flavor anf and milk is developed by the combination of EGC-NPs and whey protein?WP?since EGC-NPs possesses anti-arthritis effect with non-toxicity.The results of food hygiene evaluation and detection showed that the heavy metal pollutant index and total bacterial count index of the anti-arthritis food met the national standards.
