Perilla frutescens alias red perilla,fragrant perilla,red perilla,is the lip-shaped family perilla annual herbs,is the traditional Chinese medicine food,oil crops.China is rich in perilla resources,and the effective substances extracted from perilla leaves have significant utilization value in medicine,food,health products,daily necessities and other fields.In this paper,the anti-allergic activity of 24 perilla varieties in Shanxi Province was screened,and the anti-allergic components in perilla leaves were extracted,purified,structural identification and anti-allergic activity test,which provided theoretical basis and technical support for the development and utilization of perilla.The main findings are as follows:(1)24 perilla varieties were extracted by hot reflux extraction and spray-dried,and the samples were tested by hyaluronidase inhibition test.The results showed that variety No.22 collected from Zuoquan County,Shanxi Province had the best antiallergic ability,the inhibition rate of hyaluronidase was 49.72%(C=5 mg/mL),and the yield of extract was5.89%.(2)The effects of heat reflux method,ultrasonic extraction method,microwave extraction method,enhanced mass transfer method and cold leaching method on the content of flavonoids and polyphenols in perilla leaf extract were compared.The optimum technological conditions of perilla's antiallergic active substances and chemical components were obtained by single factor experiment with flavonoids,polyphenol content and antiallergic ability: temperature 36?,rotational speed 11000 r/min,ratio 1:15,extraction time5 min,extraction times 3 The antiallergic activity of perilla leaves was the best,and the inhibition rate of hyaluronidase was 54.35%(C=5 mg/mL)under this condition,the contents of flavonoids and polyphenols in the extract reached 46.09% and 14.12%,respectively.(3)Silica gel column chromatography and dextran gel column chromatography were selected to separate and purify the crude extract of perilla,and the antiallergic components of perilla after purification were analyzed by LC-MS,and the antiallergic activity of perilla wasdetected by SDS-PAGE method in vitro.The results showed that the inhibition rate of purified component hyaluronidase was 89.76%(C=5 mg/mL),the main antiallergic active substances in perilla were luteolin and rosmarinic acid,and it was found that the antiallergic active substances in perilla could complex with astaxanthin stably and reduce the possibility of allergic reaction...
