Cyantraniliprole was the second generation of anthranilic diamide insecticide,and triflumezopyrim was a noval mesoionic insecticide.At present,cyantraniliprole and triflumezopyrim have been registered in various vegetables and crops in China,with a wide range of applications,but little research has been done on ecological toxicity of soil organisms.In this experiment,Eisenia fetida was selected as the test organism,and the acute toxicity and chronic toxicity effects of cyantraniliprole and triflumezopyrim on Eisenia fetida were determined and evaluated according to OECD test methods.In the chronic toxicity test,the concentrations of cyantraniliprole and triflumezopyrim were all set at 0,0.5,1,2.5,5 and 10 mg/kg artificial soil,and samples were taken from each concentration treatment at 7,14,21 and 28 days after exposure respectively for experimental determination.The test indexes include reactive oxygen species(ROS),Superoxide dismutase(SOD),Catalase(CAT),Glutathione Stransferases(GST),Malondialdehyde(MDA)and 8-hydroxy-2'-deoxyguanosine(8-OHdG).Through the chronic toxicity treatment of the two insecticides on earthworms,the effects of different doses of cyantraniliprole and triflumezopyrim on the antioxidant enzyme activity and DNA damage of Eisenia fetida were studied,so as to evaluate the ecotoxicological effects of the two insecticides on earthworms and provide effective basis for their reasonable field application and environmental safety evaluation in the future.The main results of this study were as follows:(1)The acute toxicity of cyantraniliprole and triflumezopyrim to Eisenia fetida for 14 days were low.(2)Under various doses(0,0.5,1,2.5,5 and 10 mg/kg)of cyantraniliprole,compared with the control group,ROS content showed a more obvious trend of increasing with the increase of concentration in the late stage(28d)of treatment,but changed greatly in the early stage of treatment;SOD activity increased with the increase of dose and the passage of time.The overall change trend of CAT activity was not obvious,but decreased significantly at 28 d compared with the control group;The GST activity in earthworms showed a significant trend of change,first increasing and then decreasing,which played an anti-oxidation and detoxification function;MDA content in the highest concentration treatment group(10 mg/kg)showed a trend of gradual accumulation over time;The content of 8-OHdG in earthworm gradually increased,showing a dose-effect relationship with the concentration of cyantraniliprole.(3)Under various doses(0,0.5,1,2.5,5 and 10 mg/kg)of triflumezopyrim,compared with the control group,ROS had no obvious change at the early stage of treatment,and had a significant dose-effect with triflumezopyrim concentration at the late stage of treatment,and ROS content increased obviously with the increase of concentration;SOD and CAT activities were obviously induced and activated,but at 28 d,CAT activities of all treatment groups were restored to the level of control group;GST activity showed an obvious trend of increasing first and then decreasing.MDA content in the 5 mg/kg treatment group reached the highest at 21 days;The content of 8-OHdG in earthworm was lower than that in the control group at the early stage of treatment,but it showed a trend of increasing with the concentration of triflumezopyrim at 28 days,with a significant dose-effect relationship.(4)Two-way ANOVA showed that after earthworms were treated with cyantraniliprole and triflumezopyrim alone,the changes of various determination indexes in vivo were the result of the combined action of treatment dose and time,and different treatment dose and treatment time had significant effects on various biological indexes in earthworms;The results of comprehensive toxicity evaluation showed that the comprehensive toxicity of cyantraniliprole treatment group decreased gradually with the extension of time;The comprehensive toxicity of triflumezopyrim treatment group showed a significant trend of increasing first and then decreasing;Combined with RDA results,the levels of ROS,SOD,GST and 8-OHdG had a significant dose-effect relationship with the concentrations of the two agents;There was a strong correlation between SOD and CAT,ROS and 8-OHdG in the treatment of cyantraniliprole;In triflumezopyrim treatment,there was a strong correlation between ROS,SOD and GST.
