Lycopene is a high-quality antioxidant natural pigment,which has a good scavenging effect on a variety of oxidizing substances.Because of its huge potential in the field of food and medicine,it has become a hot research direction in related fields in recent years.Neurospora crassa is a fast-growing,low-nutrient model organism that is widely distributed in nature.Its excellent growth characteristics are suitable for fermentation of a variety of products.It can produce industrial and daily substances including ethanol and lycopene and has not much difference with the properties of naturally occurring substances.In recent years,it has become a hot research object.1.The research screened the types of nitrogen sources,determined the use of soybean peptone as the nitrogen source,and studied the effect of soybean peptone on the lycopene production of Neurospora crassa.In the end,the best conditions for the three factors of soybean peptone addition,addition time and inoculation amount were:addition amount 11 g/L,addition time 53 h,inoculation amount 6%,and lycopene yield reached 9.64 mg/L.2.In order to obtain lycopene extract with higher purity,the extraction and purification process of lycopene was optimized.The optimization was mainly from three aspects:material-liquid ratio,sonication time,and dark room extraction time.The order of the three factors affecting the lycopene extraction amount was:dark room extraction time>extraction liquid volume ratio>ultrasonic time.The optimal process conditions are:the volume ratio of the extraction solution is 2:3,the ultrasonic time is 20 min,and the dark room extraction time is 2.5 h.Under these conditions,the extraction amount of lycopene was 2.41 mg/g.3.Test the antioxidant capacity of the extracted lycopene crude extract and compare it with ?-carotenoids,BHT,VC,and VE,by removing O2-·radicals,·OH-radicals,DPPH · free Based on the total antioxidant FRAP method.The experimental results show that within the experimental concentration range,the ability to remove DPPH·is ?-carotenoids>BHT>VC>Lycopene crude extract>VE;O2-·has the clearing ability of Product>?-carotenoids>VC>VE>BHT;The order of removal and OH-capacity is:?-carotene>Lycopene crude extract>BHT>VC>VE;The order of total reducing capacity is:Carotene>VC>BHT>Lycopene crude extract>VE.
