With the promotion of the international "One Belt And One Road" strategy,China's food trade position is increasingly prominent,food safety is the main trade fortress.China is a big meat producer and an important field in the "One Belt And One Road" strategy.In recent years,the State Administration for Market Regulation has issued a notice on a number of incidents in which the illegal use of veterinary drugs resulted in the high residue of veterinary drugs in meat exceeding the standards.The 13th Five-Year Plan has put forward requirements on the comprehensive treatment of veterinary drug residue safety,which has aroused wide concern from the society.With the continuous update of scientific methods,it has become a difficult point for animal residue detection to construct a miniature and environmentally friendly sample pretreatment method and a high-sensitivity simultaneous detection technology for meat residues.The development of an efficient and convenient real-time detection system for animal residues is still the research direction of food safety detection.In this study,a nondirectional screening method for multiple veterinary drug residues was established based on mass spectrometry(MS)cleavage mechanism.Based on magnetic spe pretreatment technology,carbon nanotubes and metal organic skeleton as carbon source,was prepared by two different types of porous adsorbent-magnetic type zeolite type imidazole ester skeleton and magnetic carbon nanotubes material,build the two kinds of magnetic composite material as the adsorbent of sample pretreatment method,this method is environment friendly,strong selectivity,the advantages of the recycled,significantly improve the efficiency of the veterinary drug residue detection method,realized the meat in the enrichment of residual more efficient and rapid purification and accurate detection,provide technical support for food safety regulation.The main contents of this paper are as follows:In the first part:the safety status and detection methods of veterinary drug residues in meat were reviewed,and the development of magnetic solid phase extraction pretreatment technology for veterinary drug analysis was reviewed.The second part:According to the maximum residue limits of common veterinary drugs prescribed by the European Union,the United States,the Codex Alimentarius Commission,Japan and China,59 common veterinary drugs in 12 categories were selected as the target substances.Based on the study of the fracture mechanism of high resolution mass spectrometry,an analytical method for non-directed screening of 59 veterinary drug residues was constructed.Experiment with Full scan data dependent on screening(Full MS/dd-MS2)mode establishing database of veterinary drug standard,adopt not rely on data acquisition mode(Full MS/vDIA)mass spectrometry determination of retention time,accurate mass number based on standard material,secondary ions,qualitative screening information,through analyzing the mass spectrum of molecular ion fragmentation process and cracking mechanism,12 classes of veterinary drug characteristics fracture fragments and breaking rules to build the targeted screening method.The third part:A magnetic solid phase extraction method based on magnetic zeolite imidazolide skeleton material(Fe3O4@ZIF-8)was established to extract 7 sulfanilamide from mutton successfully.Several factors influencing the extraction effect were optimized,hydrophobic interaction and hydrogen bonding between SAs and adsorbants,the pore diameter of Fe3O4@ZIF-8 has excellent matching effect with SAs to promote SAs enrichment.Under the optimal conditions,a method of high sensitivity and selectivity for sulfa was established.The fourth part:A method for the rapid screening of 12 kinds of veterinary drug residues in mutton was established by using magnetic carbon nanotube adsorption material(Fe3O4@O-MWCNTs)combined with the technique of electrostatic field track trap.The magneto-solid phase extraction conditions and methodology parameters were optimized to verify that magnetic carbon nanotubes had better purification effect on meat,and the method was successfully applied to the detection of 12 veterinary drugs in mutton.
