Cyanide ion?CN-?is an enzyme inhibitor that invades the human body through the respiratory tract,skin,etc.,thereby affecting various functions,including blood vessels,visual system,central nervous system,heart,endocrine,and metabolic system.Hypochlorite ion?ClO-?,as one of the reactive oxygen species,plays an important role in the process of immunity against microbial infection and inflammation.The imbalance of ClO-concentration in life will induce the occurrence of various diseases,such as atherosclerosis,arthritis and tumors.As a kind of toxic heavy metal ions,Hg2+ is enriched into life through the food chain,which not only harms the central nervous system,but also causes gastrointestinal ulcer,respiratory failure and even death.Fluorescent probes have the advantages of high sensitivity,easy operation,in-situ and visual detection,and can be directly used for in-situ real-time detection of ions in biological processes.Therefore,the development of fast,efficient and reliable significance for environmental monitoring,food safety and medical diagnosis.In this thesis,the bifunctional fluorescent probe 2TM based on bithiophene-Miehler acid,the new fluorescent“turn-on”probe 2TS based on bithiophene and the new fluorescent probe 2TD based on bithiophene-diaminomaleonitrile were synthesized,And its structure was characterized by instrumental means.Using modern optical instruments,the optical properties of the probe such as sensitivity,selectivity,anti-interference,time response,p H sensitivity and concentration linear relationship were systematically studied,and through molecular simulation calculations and optical titration.Curves,Job's Plot curves,in situ nuclear magnetic,infrared spectroscopy,and high-resolution mass spectrometry have been used to study their identification and response mechanisms.Finally,the three fluorescent probes are used in environmental samples,food quantitative detection and in vivo cell imaging applications.Based on the above research,the following results were obtained:?1?A dual-functional high-efficiency fluorescent probe 2TM based on bithiophene-Michleric acid was developed for CN-in DMSO/H2O?1/3,V/V?solution and ClO in?100%aqueous solution.Demonstrates fast,colorimetric and fluorescent turn-on response characteristics,with specific selectivity and high sensitivity.The prepared probe 2TM test paper strip and the loaded silica powder realize the rapid on-site detection of CN-and ClO-.In addition,the probe has successfully achieved quantitative trace detection of CN-and ClO-in environmental water samples and imaging applications of CN-and ClO-in living cells.?2?A new fluorescent"turn-on"probe 2TS based on bithiophene was developed solubility,high selectivity,high sensitivity,wide p H response range,fast time response realizes rapid on-site detection of Hg2+.Importantly,the probe not only successfully biological samples,but also enables Hg2+imaging applications in living cells.?3?A new fluorescent probe 2TD based on bithiophene-diaminomaleonitrile was designed and prepared,which was used to detect ClO-in?100%aqueous medium.Probe 2TD exhibits high selectivity,high sensitivity,strong anti-interference,wide p H response range,ultra-fast time response and good biocompatibility for ClO-detection.The prepared probe 2TD test strip quickly realized the on-site detection of ClO-.In addition,the probe 2TD has successfully achieved quantitative detection of ClO-in environmental water samples and fluorescence imaging applications of ClO- in living cells.
