Cordyceps sinensis, as a medical fungi, is value of medicine. In recent years, the demand for cordyceps in domestic market increases, however, natural cordyceps needs parasitic strict and harsh living conditions, the output of the natural cordyceps is far from meeting market demand. Recent studies show that mycelium obtained by cultured cordyceps fungus,which active ingredients almost as same as that of the natural cordyceps .It can be used to making pharmaceutical and health care products, so it is important significance of cultivating cordyceps research.In this experiment a cardceps fungi E1 is obtained from Muli County of Liangshan and identification is Hirsutella sp. The optimum culture medium and fermentation conditions was determined by single factor experiment and orthogonal test.Using mannitol and cordycepin as indicators determine the optimum culture medium formula: yeast powder 1%, beef extract 2%, peptone 1%, soybean cake powder 4%,sucrose 2%, potassium dihydrogen phosphate0.15%, magnesium sulfate 1% and matrix buckwheat: optimization of fermentation process for liquid seed shaking culture time is 3 days, 10% of the volume of liquid seed inoculation, medium load 60g, moisture content is 60%, solid state fermentation time for 10 days. The content of cordycepin and mannitol was improved after optimization of culture medium and fermentation conditions, which indicated that the optimization of fermentation conditions could effectively improve the content of Codyceps sinensis and mannitol. |
