To study the effect of different pH on the gene expression of Saccharomyces cerevisiae,Pach medium and wort medium were selected to carry out aerobic culture under pH3.0?4.0?4.5?5.0?5.5?6.0?6.5?7.0?8.0?9.0 conditions in 5 L fermenter,and the transcriptome of Saccharomyces cerevisiae under different conditions was analyzed by sequencing.The results showed that there were 1416,1363,2026,3324,1925,3025,3159,3626,3104 genes differentially expressed between pH3.0 and other pH in the Pach medium.Annotated to 1654,1817,1544,1993,2259,2162,1619,2392,2113 functions inthe GO database.At KEGG database,485,532,712,1217,795,1171,955,899,1111 metabolic pathways were located.There wer e 1658,1824,1547,2003,2268,2169,1621,2405,2118 genes different ially expressed between pH3.0 and other pH in the wort medium.Annot ated to 1654,1817,1544,1993,2259,2162,1619,2392,2113 functions inthe GO database.At KEGG database,612,673,575,695,830,794,504,844,705 metabolic pathways were located.Most of the gene expre ssion of related enzymes screened in tricarboxylic acid cycle and glyoxy late cycle in aerobic metabolism decreased with the increase of pH,ind icating that the expression of this part of gene would be inhibited at hi gh pH conditions.The comparison of low pH and high pH showed that the number of differential genes in pH3:pH8?pH3:pH9 was 3104,3626,2392,2113,in Pach medium and wort medium respectively.These di fferential genes were all located in biological processes in GO enrichme nt analysis.In KEGG pathway enrichment analysis,it was found that th e genetic information transfer process and carbohydrate metabolism proc ess were mainly affected under the condition of large acid-base environ ment span.
