Development Of Eucommia Ulmoides Leaves Health Toothpaste

Nowadays people pay more and more attention to oral hygiene.A healthy mouth and fresh breath can bring pleasure to people.Toothpaste is the simplest,most convenient,and most effective oral products used for people to keep their breath fresh,maintain oral health,and clean teeth.Daily oral cleaning is an important means to prevent oral diseases and proper brushing can improve oral health.At present,there are more and more types of toothpaste,whose effect has evolved from a single oral cleaning to an effective auxiliary treatment.In order to treat oral diseases,some active ingredients are added into the toothpaste by a lot of research institutions and enterprises to achieve the effect of bacteriostasis,reducing inflammation,delaying pain and reducing plaque,etc.,which has certain auxiliary effects on periodontitis gingivitis and gingival bleeding caused by certain causes.Natural Chinese herbal medicine extracts,derived from natural herbal medicines and applied to daily chemical products with modern scientific extraction technology,is a major feature of the research and production of functional toothpaste in China.The main research contents of this paper are as follows:(1)In this experiment,the active ingredients of eucommia ulmoides leaves,extracted by ethanol extraction method,purified by activated carbon decolorization,chitosan impurity removal,and absorped by HD100 macroporous resin column adsorption,were identified by high-performance liquid chromatography.(2)In this experiment,the toothpaste process formula was optimized to determine the best formula by adjusting the ratio of thickener,moisturizer and friction agent in toothpaste and observing the changes of toothpaste appearance,stringing phenomenon,particle hardness,PH value.A pilot test was conducted in the optimized scheme with added eucommia ulmoides leaves extract to verify the feasibility of the scheme.(3)Microbial experiment and animal experiment were used to evaluate the eucommia ulmoides health toothpaste.AGAR dilution method was used to evaluate the antibacterial activity of eucommia ulmoides leaves extract in microbial experiments.The amount of addition of eucommia ulmoides leaves extract in health toothpaste was determined by the minimum inhibitory concentration of the extract against Staphylococcus aureus,Escherichia coli,Candida albicans,Proteus and Fusobacterium nucleatum.The results showed that the minimum inhibitory concentration of eucommia ulmoides extract against Staphylococcus aureus and Escherichia coli was 75g/L,and the minimum inhibitory concentration against Candida albicans showed no inhibitory effect within the experimental range.The minimum inhibitory concentration against Proteobacteria and Fusobacterium nucleatum was 50g/L.The results showed that the extract of Eucommia ulmoides leaves had antibacterial effect.The antibacterial effect of Eucommia ulmoides health toothpaste and commercial Chinese herbal medicines was compared through the bacteriostatic ring experiment.The experimental results showed that Eucommia ulmoides health toothpaste also had antibacterial activity and was higher than the commercial Chinese herbal toothpaste.The 20% Eucommia ulmoides leaf health toothpaste has a diameter of 20.3 mm for Staphylococcus aureus,11.8 mm for Escherichia coli,16.6 mm for Proteus,11.6 mm for Fusobacterium nucleatum,which indicated that Eucommia ulmoides leaves health toothpaste has bacteriostatic effect.(4)In the evaluation of animal experiments,a model of xylene-induced ear swelling and inflammation in mice was established to judge the anti-inflammatory effect of eucommia ulmoides health toothpaste,in which dexamethasone acetate ointment was used as a positive drug;at the same time,a hot plate-induced analgesia test in mice was established to judge the analgesic effect of eucommia ulmoides leaves health toothpaste,in which aspirin was used as a positive drug.In the xylene-induced mouse ear swelling experiment,the ear swelling value of the mice in the eucommia ulmoides leaves health toothpaste group containing 10% eucommia leaves extract was 4.633 ± 3.61,and the ear swelling value of the mice in the eucommia ulmoides leaves health toothpaste group containing 20% eucommia ulmoides leaves extract was 2.333±0.61.The ear swelling value of mice in the positive drug group is 3.533±1.23.The results showed that eucommia ulmoides leaves health toothpaste and positive drug dexamethasone acetate ointment have anti-inflammatory effects,and the anti-inflammatory effect of eucommia ulmoides health toothpaste containing 20% eucommia ulmoides leaves extract is significantly higher than that of positive drugs;In the experiment of pain induced by hot plate in mice,the health toothpaste containing 10% and 20% eucommia ulmoides leaves and the positive medicine aspirin all prolonged the pain induced by hot plate in mice.The results showed that eucommia ulmoides leaves health toothpaste also has analgesic effect.Conclusion: This study proved that eucommia ulmoides leaves extract had abroadspectrum antibacterial effects.The eucommia ulmoides leaves health toothpaste has bacteriostatic effects on oral pathogens,and it can reduce the swelling rate of xylene-induced ear inflammation and relieve pain in mice.Animal evaluation showed that eucommia ulmoides health toothpaste had anti-inflammatory,antibacterial and analgesic effects.
