Fruit mulberry is a kind of plant that can be used by the whole plant and is "medicine-food homologous".Its fruits,leaves,branches,and roots are rich in nutrients and active ingredients,which has the function of cleansing the lungs and moistening dryness,nourishing the liver and improving eyesight,anti-oxidation,lowering blood sugar and blood lipids and enhancing immunity,and so on,has broad prospects for development.However,the current development focus of mulberry is mainly on mulberry fruit.Insufficient use of mulberry leaves,mulberry branches and mulberry roots has caused waste of fruit mulberry resources.In this paper,mulberry leaves,mulberry bark and mulberry root bark are used as raw materials,and the design of the mixture is used.The optimal ingredient of the mulberry mixed tea is determined by taking the sugar-reducing active ingredients(1-DNJ,flavonoids,and polysaccharides)as indicators.And optimize its brewing conditions,evaluate its hypoglycemic activity through animal experiments,and explore the effect of fruit mulberry tea on the intestinal flora of type 2 diabetes mellitus model mice.The new fruit mulberry tea product with nutritional value aims to provide an effective way to improve the comprehensive utilization value of fruit mulberry resources.The main research contents and results are as follows:1.Analysis of nutritional and functional components of mulberry leaves,mulberry bark and mulberry bark.Mulberry leaves contain higher crude protein,ash and crude fat,and mulberry bark and mulberry bark contain higher crude fibers.All three kinds of raw materials contain a certain amount of amino acids,and the essential amino acids are complete,so the nutritional value is good.In terms of functional ingredients,the content of flavonoids in mulberry leaves is the highest,which is 50.84 mg / g,and the content of polyphenols and polysaccharides in mulberry bark are the highest,which are 59.85 mg / g and 4.02 g / 100 g,respectively;the content of 1-DNJ in mulberry bark is the highest,which was 2.714 mg / g.2.Study on the preparation of fruit mulberry tea.Using the dissolution rate of polyphenols,flavonoids,polysaccharides,and 1-DNJ as indicators,the optimal crushed meshes of mulberry leaves,mulberry bark,and mulberry root bark were determined to be 40 to 60 mesh,60 to 80 mesh,and 60 to 80,respectively.Using flavonoids,polysaccharides,and 1-DNJ as responsevalues,using blend design,sensory evaluation,and inhibition rate of ?-glucosidase to obtain the optimal blending ratio of fruit and mulberry tea: fruit and mulberry: mulberry Branch bark:mulberry bark is 62:19:19;the optimal brewing conditions are obtained by orthogonal test as fllows: brewing water temperature was 75 ?,tea-to-water ratio was 1: 100,brewing time was 50 min.3.Study on the hypoglycemic effect of fruit mulberry tea.In vitro hypoglycemic studies have shown that the inhibitory rates of ?-glucosidase and ?-amylase by mulberry tea infusion solution are 74.77% and 68.04%,respectively.Based on this,a mouse model of type 2 diabetes mellitus was constructed.Set GD-1 group(including 5% mulberry compound tea high fat feed),GD-2group(including 10% mulberry compound tea high fat feed),GD-3 group(mulberry compound tea extract 200 mg / kg)three groups of tests,with metformin(100 mg / kg)as a positive control,continuous drug delivery for 28 days,determination of food intake,water consumption,fasting blood glucose(FBG),serum insulin(INS),glycated serum protein(GSP),glycated hemoglobin(GHb A1c),and oral glucose tolerance(OGTT)and other biochemical indicators to evaluate the hypoglycemic effect of fruit mulberry tea,the results showed that: compared with the normal group,the fasting blood glucose level of the model group of mic was always at a high blood glucose level,and the water consumption was significantly higher than the normal group;the positive drug group in the administration group had the strongest ability to lower blood sugar,and the best hypoglycemic effect among the three groups of fruit mulberry tea with different dosages was the GD-2 group,which had an effect on fasting blood glucose(FBG),the decrease of GSP and glycosylated hemoglobin(GHb A1c)were 40.05%,23.28%,and 15.73%,respectively,and there were no significant differences compared with the positive control;the GD-1 and GD-3groups had a certain degree effect to reduce the blood sugar,but not as effective as the GD-2group.4.Application of high-throughput sequencing technology for mouse intestinal flora analysis.The results showed that the composition of the intestinal flora of diabetic mice was significantly different compared with other groups,and the specific manifestations were increased abundance of harmful bacteria such as Bilophila and Clostridium,and beneficial bacteria such as Akkermansia.The abundance of the genus decreased;the results of Alpha diversity analysis showed that the intestinal flora diversity index of type 2 diabetic mice was lower than that of thenormal group,and the rest groups were higher than the model group in different degrees,of which the GDm group(containing 10% fruit and mulberry tea high-fat diet)was the highest;the results of Beta diversity analysis showed that the species composition of the intestinal flora of the model group mice was the same as that of the other groups The differences were found through PCA analysis and UPGM analysis.The intestinal species composition of mice in the GDm group and the normal group was similar to a certain extent,indicating that the fruit mulberry tea has a certain role in regulating the intestinal species composition of diabetic mice toward the normal group.The LEf Se multi-level species discriminant analysis results show that the bacteria that have a greater effect on the intestinal flora of the model group are harmful bacteria such as Bilophila,and the bacteria that have a greater effect on the remaining groups are Bacteroides.Fungus.The above results indicate that fruit mulberry tea had a regulating effect on the intestinal flora of diabetic mice,which was manifested in the composition and structure of the flora to improve the symptoms of intestinal flora imbalance in diabetic mice.
