Wampee(Clausena lansium)is a kind of excellent and rare fruit in south China,rich in polyphenols and other nutrients.In this study,158 varieties of wampee germplasm resources were used as samples to explore the polyphenol composition,total phenolic content and in vitro antioxidant activity in the fruits of different varieties,different development stages and different parts,and to preliminarily investigate their lipid-lowering activity.The main results are as follows:(1)The total phenolic content of extracts from 158 wampee germplasm resources fluctuated greatly due to the diversity of the varieties,ranging from 49.25 to 1055.21 mg GAE/100 g FW.Seven phenolic components(syringin,2-methoxycinnamic acid,hesperitin,nobiletin,tangeretin,rutin and kaempferol)were detected from the extracts.The total antioxidant capacity(ORAC)varied widely,from 0.64 to 13.92 mmol TE/100 g FW.The cellular antioxidant capacity(CAA)also fluctuated greatly,among which the No wash value varied from 7.51 to 941.49 mol QE/100 g FW,and the Wash value varied from 0.83 to 248.68 mol QE/100 g FW.The average total phenolic content of wampee extract was higher than that of common fruits,and the antioxidant activity in vitro was generally higher than that of most citus fruits.Correlation analysis showed that there was a significant positive correlation between total phenolic content and ORAC value(p<0.01).In addition,principal component analysis simplified 8 evaluation indexes of germplasm resources s into 4 principal components.The evaluation indexes of 11 germplasm resources s were preliminarily classified into 6 categories and 158 germplasm resources into 10 categories by hierarchical cluster analysis.(2)CL-132 and CL-133 were selected from two wampee germplasm resources with significant difference in total phenolic content and in vitro antioxidant activities.During three stages of wampee fruit development,it was found that the total phenolic content decreased significantly.Also,the the phenolic monomers content continued to decrease with different degrees.The soluble sugar components(fructose,glucose and sucrose)were continuously accumulated but the overall organic acid level decreased.Moreover,in vitro antioxidant activity was significantly declined.The absorption rate of antioxidant components of Hep G2 cells showed an opposite trend in two varieties.The cytotoxicity to Hep G2 cells was enhanced to some extent during different stages and the change trend of anti-proliferative activity against Hep G2 cells was different among varieties.(3)The pulp and peel of five wampee germplasm resources(CL-2-1,CL-40,CL-113,CL-154,CL-164)with total phenolic content and antioxidant activity at different levels were selected separately as samples.It was found that the total phenolic cotent and antioxidant activity in vitro were higher in peel extracts than that in pulp extracts.Fatty liver model was established by 0.5 mmol/L oleic acid-induced Hep G2 cells,and oil red O staining showed that 12.0 mg/m L CL-154 peel extract had the best lipid-lowering activity.The results showed that wampee fruit as medicine and food,had higher total phenolic content and in vitro antioxidant activity than other common fruits.In terms of fruit development,CL-132 was similar to that of acidless citrus model,and CL-133 was similar to that of mediumacidic citrus model.In addition,as a potential lipid-lowering active material,wampee peel has higher application value in the development of functional foods.
