Betel nut,as one of the four southern Chinese medicines,is rich in active ingredients,which has the function of eliminating food,removing insects and protecting health.The physiological effects of arecoline are gradually clarified,but the physiological effects of arecoline polyphenols are rarely reported.It is pointed out that the content of phenols in areca seed is the highest in all parts of areca nut,and about 90% of plant polyphenols need to be metabolized by intestinal flora before they can be absorbed and utilized by human body.In this experiment,areca seed polyphenols was extracted and separated,and the culture technique of human intestinal bacteria in vitro was applied to the interaction between the intestinal flora of the stripling and the areca seed polyphenols in vitro.It will be explained that the effect of areca seed polyphenols on human health by comparing with green tea polyphenols to explore the biological transformation of areca seed polyphenols,and the change in the diversity of human intestinal flora and the content of metabolites.The main results are as follows:1.Polyphenols in areca seed were extracted by organic solvent extraction method,and the extraction rate of polyphenols in areca seed was up to 40.43% by optimizing acetone volume fraction,extraction temperature,extraction time and solid-liquid ratio parameters.2.XAD-7HP macroporous resin was used to separate and purify the polyphenols of areca seed,and the purity of the polyphenols of areca seed could be improved from 44.03% to 85.96% after the screening and optimization of the process parameters of static adsorption-desorption and dynamic adsorption-desorption.3.In the experiment of betelnut seed polyphenols and green tea polyphenols fermented by human intestinal flora,it was found that there were significant differences between the two polyphenols,which led to differences in the absorption and utilization of the intestinal flora of young men.In terms of the utilization of polyphenols,the total phenols and catechins in the green tea group were more useful than those in the betel nut group,and the accumulation of SCFAs in the betel nut group showed a stronger promoting effect.4.The addition of betelnut seed polyphenols and green tea polyphenols is the main cause of microbial changes during fermentation,and the effect of betelnut seed polyphenols on intestinal flora is stronger than that of green tea polyphenols at the same total phenol concentration.Areca seed polyphenols can significantly increase species abundance and flora correlation,but increase the abundance of some pathogens,and the effect of green tea polyphenols is significantly different.In summary,areca seed polyphenols can affect the structure and metabolism of human intestinal flora,regulate the pH of intestinal environment,and promote the metabolism of human intestinal flora to generate more SCFAs,but at the same time increase the abundance of some pathogenic bacteria in the intestinal flora.Compared with green tea polyphenols,the effect of areca seed polyphenols on intestinal flora is more obvious,which may be related to the difference in composition between the two groups.
