Abstract/Summary:
China was the world’s largest producer and consumer of cauliflower.The edible part of cauliflower composed of flower buds,flower branches and flower axles which made ball-flower,but a large of stems and leaves were unused.Glucosinolates were important secondary sulfur-containing metabolites in cruciferous plants,which had anti-cancer,anti-inflammatory,anti-oxidation and other biological activities.In this study,it took cauliflower leaves as raw material,adopting orthogonal test to optimize the extraction conditions of glucosinolates and identifing the kinds of different varieties of cauliflower leaves’glucosinolate by HPLC-Q-TOF-MS technology.It provided scientific thinking for resource utilization of glucosinolate and scientific basis for rapid identification of glucosinolate.1.A comparative research of glucosinolates’extraction in varies of cauliflower leavesThe experimental factor such as extraction time,ethanol concentration,liquid to solid ratio and extraction temperature were studied with the content of glucosinolates as the index and investigated by single factor and orthogonal design.Study have shown that:The Baota cauliflower:the best extraction time was 50 min,the best ethanol concentration was 90%,the best liquid to solid ratio was 1:10(W/V)and the best extraction temperature was 90 ~0C.On the basis of this condition,The Baota cauliflower leaves had content of glucosinolates reaching 94.89μmol/g;The Qinghua cauliflower:the best extraction time was 40 min,the best ethanol concentration was 80%,the best liquid to solid ratio was 1:14(W/V)and the best extraction temperature was 80 ~0C.On the basis of this condition,The Qinghua cauliflower cauliflower leaves had content of glucosinolates reaching 92.51μmol/g;The Zise cauliflower:the best extraction time was 40 min,the best ethanol concentration was 80%,the best liquid to solid ratio was 1:14(W/V)and the best extraction temperature was 80 ~0C.On the basis of this condition,The Zise cauliflower cauliflower leaves had content of glucosinolates reaching 112.75μmol/g.2.Identification of glucosinolate in leaves of different varieties of cauliflowerA method for analysis of glucosinolates in three kinds of cruciferous vegetable leaves was developed by using HPLC-Q-TOF-MS.Firstly,the mass spectrometry fragmentation pathways of two standard glucosinolates was studied.The bond of glucose,sulfated thiohydroximate and R-group was easy fractured and the rearrangement reaction was observed for the forming the fragment ions.Then,the structure of 9 glucosinolates including3-(Methylsulfinyl)propyl(1),4-(Methylsulfinyl)butyl(2),3-Butenyl(3),4-Hydroxybenzyl(4),2-Propeny(5),1-Hydroxyindol-3-ylmethyl(6),Indol-3-ylmethyl(7),1-Methoxyindol-3-ylmethyl(8),6-(Methylsulfonyl)hexyl glucosinolate(9).There are 4 kinds of glucosinolate(1-2,4,6)in Baota Broccoli,6 kinds of glucosinolate(2-3,6-9)in Qinghuacai and 7 kinds of glucosinolate(1-2,4-8)in Zise Broccoli among these glucosinolate.
