Cancer,one of the most recognized diseases in the world,kills about 9 million patients every year.It involves abnormal growth of human cells and is easily invaded or spread to other parts of the body.The research for cancer healing has not yet been successful.Clinically,chemotherapy,hormone therapy,surgery and other treatments are commonly used to change their cellular DNA fragments to inhibit the disease and limit the rapid division of cancer cells.Among them,platinum-based anticancer agents,cisplatin and its derivatives,carboplatin,oxaliplatin,nedaplatin and other drugs have a good therapeutic effect.However,the use of these drugs is accompanied by other damage to the body of cancer patients,such as nerve damage,hair loss and other symptoms,and even shows resistance to cancer cells.Therefore,people are eager to design and synthesize anticancer drugs with novel structures,anti-tumor effects and low toxic side effects.In recent years,lanthanide complexes have been extensively studied due to the structural diversity and their unique medical value.In this paper,two kinds of metal complexes were prepared using carboxylic acid ligands with abundant conjugated elements and their antitumor activities were studied.The main work contents are as follows:1.Select and two compounds are organic ligands,and two metals are prepared by controlling hydrothermal synthesis reaction conditions with cerium nitrate and cerium nitrate,respectively.Complex 1(C44H22N2O18Sm2)and 2(C30H20ErO8)were prepared by solvothermal method with organic linkers of 2'-amino-[1,1':4',1"-terphenyl]-3,3",5,5"-tetracarboxylic acid?L1?and [1,1':4',1??-terphenyl]-4,4??-dicarboxylic acid?L2?.The complexes 1-2 were characterized by infrared spectroscopy and elemental analysis,and the crystal structure of the complexes were determined by X-ray single crystal diffractometer;2.UV-spectroscopy and fluorescence spectroscopy were used to prove that complexes 1-2 were bound to calf thymus DNA?CT-DNA?by insertion mode,and the UV bonding constants of complexes 1-2 and CT-DNA were calculated.Besides,tern's constant,and comparative binding strength of complexes 1-2 with DNA were analysied;3.The application of electrophoresis gel technology proves that complexes 1-2 have good cleavage ability to plasmid pBR 322 DNA,and molecular docking proves that complexes 1-2 can be embedded in DNA molecules with specific functions;4.The ability of complexes 1-2 to induce apoptosis of cervical cancer cells?HeLa cells?was determined by Annexin V-FITC and PI double staining.The experimental results show that complex 1-2 can effectively induce HeLa cell apoptosis.The inhibition rate of Hela cells by complexes 1-2 was determined by colorimetry and compared with cisplatin.
