In order to develop drug loaded composite nanomaterials with good biocompatibility and sustained-release function,and easy to monitor the drug's operation and distribution in vivo,this project uses hydroxypropyl cellulose(HPC)to prepare graphene oxide(GO)is modified to reduce the cytotoxicity of GO,increase the drug loading,increase the absorption rate of anti-tumor drugs,improve the antitumor effect,and make the drug carrier have fluorescence,which is convenient for drug tracing.Firstly,two hydroxypropyl cellulose modified graphene oxide materials were synthesized: hydroxypropyl cellulose-graphite oxide(HPC-GO)and hydroxypropyl cellulose-Graphene oxide quantum dots(HPC-GOQDs).It was characterized by fluorescence observation,FTIR,XPS,1H NMR,TG and SEM.Characterization results show that hydroxypropyl cellulose modified graphene oxide has been successfully synthesized.The results of fluorescence observation show that both HPC-GO and HPC-GOQDs have strong fluorescence.TG analysis showed that the thermal stability of HPC-GO was better than that of HPC-GOQDs.At the same time,the toxicology of the new materials was studyed: the survival rate,body length,body width and pregnancy of nematodes were determined by culture of Caenorhabditis elegans.The results showed that compared with GO,the toxicity of HPC-GO and HPC-GOQDs decreased significantly,and there is no significant difference in toxicity between them.Then,HPC-GO nano-hydrogel beads and HPC-GOQDs nano-hydrogel beads were prepared by physical crosslinking of HPC-GO and HPC-GOQDs with Fe Cl3·6H2O.They were used as drug delivery systems to control the release of doxorubicin(DOX).The experimental results show that,HPC-GO has better swelling ability and drug loading than HPC-GOQDs,but HPC-GOQDs has a better sustainedrelease effect.Finally,cancer cells were used for experimental verification,the experimental results show that even at relatively high concentrations,no obvious cytotoxicity was observed in HPC-GO nano-hydrogel beads and HPC-GOQDs nano-hydrogel beads without DOX loading,and the cytotoxicity of HPC-GOQDs was lower than that of HPC-GO.When DOX was loaded,they were more lethal to human colon cancer cell SW480,and the lethality of HPC-GOQDs/DOX was greater than that of HPCGO/DOX.It can be seen that HPC-GO/DOX and HPC-GOQDs/DOX have good cancer treatment efficacy.
