Biomass is the most logical carbon-based feedstock obtained from living organisms such as plants,animals,and microorganisms.Among biomass,lignocellulose biomass is the most common,which is composed of various polysaccharide celluloses,hemicelluloses,phenolaldehyde polymer lignin,and soluble polar and non-polar substances.It could be conversed to the fermentable sugars though biomass transformation.However,the pretreatment and enzymatic hydrolysis are inhibited by the complexity of its internal structure,which inhabits its further conversion.Therefore,it is necessary to develop the co-effective pretreatment technology to break the barriers of lignocellucose.Hence,in this study,sugarcane bagasse and corn cob residue are used as materials to improve the process of pretreatment.The results are has follows:Firstly,the single factor experiments of sugarcane bagasse pretreated by KOH were studied.The optimum conditions obtained by component analysis and enzymatic saccharification were the KOH dosage of 15%(to oven dry stock,the same below),90 °C,solid-liquid ratio of 1:10 and reaction time of 2 h.And then the enzymatic saccharification experiments of different process were carried out to observe the change rule of sugar yield.It was found that the glucose concentration could reach up to 47.62±1.06 g/L under the optimal conditions.Morever,the total sugar yield was 81.6±1.1% under the condition of 5% solid content,20 FPU cellulase/g-substrate.Secondly,the KOH pretreatment of corn cob residue was investigated based on the preliminary research of the laboratory,the optimal pretreatment conditions were 16% KOH,70 °C solid-liquid ratio 1:10 at 90 min.Glucose yield as the function of enzymatic hydrolysis time with different solid loading and enzyme loading was explored,it was found that the glucose yield of 15% solid content,20FPU/g-substrate was 87.4%.The efficiency of enzymatic saccharification with different enzyme loading under 20% solid loading was investigated by two stage enzymatic hydrolysis.Results performed that the glucose yield was up to 91%under 20% solid loading and 20 FPU cellulase/g-substrate.At last,the comparison of the overall process and mass balance of KOH pretreatment and enzymatic hydrolysis of sugarcane bagasse and corn cob residue was carried out.Results showed that the fermentable sugar which can be used to biomass refining after enzymatic saccharification of 1 kg corn cob residue pretreated by KOH was 116.1 g more than sugarcane bagasse.
