Last several years,ferulic acid has become a minion in the field of functional food,medicine and cosmetics.Its good properties of free radical scavenging,anti-bacterial and anti-inflammatory,cancer prevention and blood lipid reduction are deeply loved and concerned by people,so it has a good prospect of development and application.China is a large country of wheat production.Wheat bran,as a by-product of wheat,is mostly used as animal feed directly,and rarely used for further processing and reuse,which results in a great waste of resources.Wheat bran is used as raw material in this project.Extraction of ferulic acid from bran by solid state fermentation.And the glucose group with strong hydrophilicity was introduced into ferulic acid.A new feed additive was prepared with ferulic acid glucose ester as the main component.The main research methods and results are as follows:The best technological conditions of ferulic acid production from wheat bran by Fusarium oxysporum.Through single factor and orthogonal test,taking the yield of ferulic acid as the index,the optimal production process parameters of ferulic acid were determined as follows:5%inoculum amount,30?culture temperature,4:3 ratio of material to liquid and 5days culture time.Under these conditions,the yield of ferulic acid was 71.45%.The ferulic acid sample obtained after concentrated freeze-drying was used as raw material.Glucose group was introduced to modify its structure.Taking the conversion rate of ferulic acid as the index,the optimum technological parameters for the synthesis of ferulic acid glucose ester were determined by single factor and orthogonal test:Using butanone and 1%methanol as reaction solvent,the reaction time was 32h,the reaction temperature was 60?,the amount of immobilized lipase was 10%,and the molar ratio of acid and sugar was 1:1.The highest conversion of ferulic acid was 25%.In order to further improve the synthesis rate and conversion rate of sugar esters,The gold enzyme hybrid system formed by gold ion and Candida lipase was used as a new catalyst to catalyze the synthesis of glucose ferulate.The changes of lipase structure before and after hybridization were determined by UV and IR,and the best technology of preparing hybrid enzyme was determined:0.7g Candida lipase was added to 10ml of 11.55nm nano gold solution,reaction in 30?constant temperature oscillating reactor for 24h.The optimum synthesis process of sugar ester catalyzed by hybrid enzyme was determined as follows:With butanone and 1%methanol as the reaction solvent,a higher ferulic acid conversion rate of36.8%can be obtained when the molar ratio of ferulic acid and sugar is 1:1,the reaction time is24 h,the reaction temperature is 60?,and the amount of immobilized lipase is 10%of the substrate.At the same time,the physical and chemical properties and biological properties of ferulic acid before and after modification were studied.It is found that glucose ester has better abilities than ferulic acid.Both of them have excellent scavenging effect on DPPH free radicals,At the concentration of 10mg/ml,the clearance rate was 64.5%and 79.8%respectively.Similarly,ferulic acid and ferulic acid glucose ester have good scavenging effects on hydroxyl radicals and H2O2.Ferulic acid and ferulic acid glucose ester have significant chelating ability to Fe2+,which are positively correlated with the concentration.At the concentration of 6.0mg/ml,the chelating ability of ferulic acid glucose ester to Fe2+is 67%,which is 1.2 times of ferulic acid at the same concentration.At the same time,compared with ferulic acid,ferulic acid glucose ester has better water solubility,which is 28 times higher than ferulic acid at room temperature.The results of bacteriostatic test showed that both of them had a certain inhibition ability to Escherichia coli,yeast and Staphylococcus aureus,and had no significant inhibition effect on Aspergillus niger.
