Androstenedione plays an important role in the field of hormone drugs development,because it can be used as an important intermediate to synthesize many other types of hormone drugs.Some relatively large amounts of androstenedione were obtained from plants in the early days,and later,androstenedione produced by microorganisms occupied the market because of its low cost and less pollution.At present,it is common in China's industry that this androstenedione is difficult to be efficiently separated from biological fermentation broth.Therefore,research on low-cost,efficient and environmentally-friendly cleaner production process solutions is imminent.In this thesis,a clean production process with simple,efficient and reasonable operation as the starting point is developed,and an extraction and separation method based on efficient extraction and separation,convenient operation,energy saving,and green safety is developed.The main results are as follows:1.The membrane treatment process was used to concentrate the moisture in the fermentation broth,and the optimal processing conditions for membrane concentration were optimized.The secondary membrane treatment method was used to remove excess moisture in the fermentation broth.The optimized membrane treatment conditions are:inlet pressure of 0.25 MPa,outlet pressure of 0.15 MPa,surface flow rate of 4.0 m / s,and temperature of 35 ?.The results show that the water removal rate of the concentrated water using the membrane can reach 74.88%.2.The emulsification properties of the system were studied,and the demulsification process plan was optimized,so that androstenedione was extracted from the emulsified system,and the extraction rate of androstenedione was determined by adding to the extract Add demulsifier and analyze whether the experiment has achieved the expected demulsification effect.3.The experimental scheme for extracting androstenedione was studied,and the androstenedione was extracted and separated by alkaline precipitation solvent complexation.Based on the single factor test,the optimal extraction condition was determined by the orthogonal test analysis method as the concentration of the methanol solution.It is 85%,pH value is 8.5,extraction temperature is 35 ° C,and the amount of demulsifier is 7%.Under these conditions,the crude androstenedione yield is 89.45%,and the purity is 85.5%.4.The experimental scheme for crystal refining of androstenedione was studied.Androstenedione was obtained using the experimental scheme of temperature increasing dissolution-evaporation concentration-cooling crystallization.On the basis of single factor test,the best crystal was determined by orthogonal test analysis method.The conditions are that the crystallization residence temperature is 17 ° C.,the crystallization residence time is 18 h,the stirring speed is 50 r / min,and the number of crystallization times is 1.Under this condition,the yield of androstenedione ketone is88.37%,and the purity is 96.5%.5.Compare the new research and development process with traditional production process.In terms of economic benefits,the annual after-tax profit and annual profit rate of the process have been greatly improved.In terms of environmental benefits,the clean production process has greatly reduced the environmental pollution.,And there has been a significant increase in energy saving and consumption reduction,the results show that the project is feasible.
