| The rapid development of biochip technology has brought great technological innovation to many fields such as life science,medicine,microbiology,chemistry and so on,and has broad application prospects.Biochip technology is the biological macromolecules such as nucleic acid fragments were orderly solidified in silicon and other solid support,and then hybridized with the target molecules in the sample to be measured by a specific device detection and analysis of fast,accurate and efficient for hybridized samples,and then determine the number of target molecules in biological samples.Manufacturers at home and abroad on the basis of biochip technology,research and development of various types of biochip detector,including temperature control system of PCR amplification and fluorescence detection system,but most of the manufacturers of biochip detector in PCR amplification stage spend a lot of time,leading to the detection process is long and low efficiency.Therefore,developing a set of rapid PCR amplification temperature control system is the key to improve the detection efficiency of biochip detector.In order to detect the effect of PCR amplification,this paper also design and build a high-precision fluorescence detection platform system.On the basis of developing a fast temperature control system,this paper puts forward the technical parameters,such as maximum rise and fall temperature speed,temperature uniformity in wind field,precision of temperature control and so on.In order to realize these technical parameters,this paper takes the Singlechip of STM32 as the lower computer and theIntelligent LCD touch screen as the upper computer to carry out the overall design of the control system.In terms of software design,the design of human-computer interaction interface,communication protocol and other modules based on JavaScript language.This method uses the software simulation of PID controller based on improved PID algorithm for temperature control,at the same time in order to meet the temperature control system can still good in different environment,this paper designs a new set of PID parameters self-tuning procedures and can be in a relatively short period of time the automatic tuning of PID control parameters for the current working environment.In terms of hardware design,this paper puts forward its own design idea according to the use occasion and designs printed circuit boards,such as main control circuit board,temperature acquisition board and stepping motor drive circuit board,and makes static and dynamic debugging of the circuit board.In this paper,a kind of heating method based on "double PWM" is proposed,and the temperature uniformity of the wind field is improved.In order to meet the requirements of the country’s medical equipment,the electromagnetic compatibility of the system was designed.The temperature control system and the principle prototype of the fluorescence detection have been completed,and the software and hardware system are debugged.After a lot of experiments and tests,the technical parameters of the temperature control system have met the design requirements.The PCR amplification effect has been well tested on the fluorescence detection platform,and verified the feasibility of the whole system. |
