Despite the continuous improvement of people's living standards,the incidence of cancer is increasing year by year.Among them,the incidence of leukemia ranks sixth,and it is also one of the major diseases with high mortality in recent years.Leukemia can be diagnosed early by hematological cytology.The basic process of leukemia is to observe blood smears containing blood samples under a microscope.Therefore,the preparation and staining of blood smears are very important for the detection of leukemia.However,at present,the preparation of blood smears mostly depends on manual operation,and the repeatability is not high.The diagnosis results are not uniform.It is easy for medical staff to misjudge the patient's condition and cause serious consequences.Therefore,the application of automated blood smear preparation and dyeing device is of great significance.At present,domestic and foreign related devices exist or are expensive,or their functions are not complete,or their efficiency is low and the process is complex.Based on the current situation,this paper studies and develops a low-cost and perfect blood cytology testing device.In this paper,the theory and application of blood cytology examination device are systematically studied.According to the different application requirements of blood smear preparation equipment,two kinds of blood cytology examination devices,high throughput and portable,are designed.The main contents include:(1)The etiology and principle of leukemia were reviewed,and the development status of blood cytology examination device was studied.The advantages and disadvantages of the existing device were compared;(2)Based on the shortcomings of high cost and imperfect automation function of existing devices,and combined with different application occasions,a design framework for developing low-cost,efficient and convenient hematological cytology inspection device is proposed;(3)A high-throughput blood cytology testing device is designed for high-sample-size testing institutions(such as large hospitals or testing centers).The relationship between pushing parameters and the effect of blood smear preparation is mainly studied.The structure design,hardware design and software design of each module are given according to the conclusions,especially the pushing mode,communication protocol and liquid level testing technology.Indepth study,and finally performance testing;(4)Aiming at low sample size testing institutions(such as community hospitals or related scientific research institutes),a microfluidic chip-based POCT(Point of Care Testing)device was designed.By analyzing the principle,material and process of making the chip,the preparation scheme of blood smear based on microfluidic chip was selected and the preparation of blood smear chip was carried out.And testing.Then,with the microfluidic chip as the core,the software and hardware design of the POCT device is completed,and its performance is tested.The performance tests of two blood cytology test devices,high throughput and POCT,show that the device runs stably,has high automation,greatly improves the quality and efficiency of push-film dyeing,and has high reliability.The device has low cost,simple and convenient operation,and achieves the expected design goals.
