Anaerobic fermentation is one of the most important methods for the utilization of distiller’s grains.The reasonable utilization of biogas residue is not only related to the smooth operation of biogas project,but also an important source of its economic benefits.At present,the utilization of biogas residue is extensive,not only low efficiency,but also secondary environmental pollution.Based on the high value utilization of biogas residue from Maotai flavor liquor distiller’s grains,the experiment of biogas residue matrix was carried out in order to obtain the feasibility of biogas residue application in tomato seedling and cultivation and the optimal matrix formula.The biological effect of biogas residue in the process of substrate was explored by 16 S rRNA high-throughput sequencing technology.It provides the application basis and theoretical basis for the high value utilization of biogas residue.The main results are as follows:1.The fermented and compounded residue of Maotai flavor distiller’s grains can be used in substrate seedling.Biogas residue is not suitable to be used as the substrate of seedling,which can inhibit the germination and emergence rate of seeds to a certain extent.After fermentation and comminution,biogas residue mixed with pit mud and vermiculite has achieved good seedling effect.The matrix formula is fine slag: vermiculite: pit mud mass ratio of 15:8:2,which can promote the growth of tomato,increase the biomass and enhance the seedling index.The correlation analysis of physical and chemical properties of substrate with plant height,stem diameter,chlorophyll and strong seedling index showed that EC value had a significant positive correlation with chlorophyll content,a significant positive correlation with stem diameter,and a significant positive correlation with pH value and strong seedling index.There was no significant correlation between bulk density and void ratio and four tomato growth indexes.2.After fermentation,the biogas residue substrate can be used in organic substrate cultivation.Compared with the conventional substrate(coconut bran,peat)tomato cultivation experiment,the biogas residue substrate can promote the growth of tomato,increase the yield of tomato,significantly improve the quality of tomato,in which VC,soluble sugar and soluble protein are significantly higher than the conventional substrate.The C / N ratio of tomato plant was highly correlated with the quality index,significantly and extremely significantly negatively correlated with soluble sugar and soluble protein,and significantly positively correlated with titratable acid.The results of principal component analysis showed that the quality of tomato treated with zh was the best,and the quality of tomato treated with YK was the worst.In conclusion,Zh treatment(fine slag: coarse slag 2:1)is superior to other three treatments in biology,yield and quality.3.There are significant differences in microbial diversity and species composition between biogas residue matrix and conventional matrix.The same substrate(ZX and zh)had the same community structure in fungi and bacteria.In terms of bacteria,the dominant species of the four substrates are the same at the gate level,but the relative abundance is different,and there are significant differences.The specific dominant phylum of biogas residue substrate is Firmicutes,accounting for 95 % of the total abundance of the species.Bacillaceae,thermoactinomycete,jg30-kf-cm45 and pseudonocardiaceae were significantly enriched,which were significantly higher(0.001 < p ≤ 0.01)than CT and YK.In fungi,Ascomycota is the dominant phylum of the four substrates,accounting for more than 70 % of all substrates,and there are significant differences with other species.The species of cepharothecaceae were significantly enriched in the biogas residue matrix.Bulk density,EC,organic matter and potassium are the environmental factors that are significantly related to the composition and abundance of microbial community.Among them,organic matter is the most important environmental factor. |
