The force that an object receives due to the attraction of the earth is called gravity.The gravity acceleration of an object in a microgravity environment generally does not exceed 10-4?.In order for astronauts to live and work normally in the space environment,they must adapt to and overcome a series of adverse effects caused by the microgravity environment,thus helping astronauts overcome a series of health problems caused by being in the space environment for a long time has become Urgent.In order to explore the influence of microgravity environment on the organism's hematopoietic system,the experimental part used zebrafish as a model animal to induce the embryos by simulating the microgravity effect,and carried out dyeing experiments to study the development of the hematopoietic system and migration of hematopoietic sites by the microgravity environment The impact of bioinformatics analysis takes human peripheral blood lymphocytes as experimental materials,simulates microgravity effects as induction conditions,and conducts subsequent research content such as gene expression profile chip experiments and differentially expressed gene screening.The development of this subject explores the impact of the microgravity environment on the hematopoietic system from the individual level and the molecular level,and provides a possible research direction for further follow-up research.The results of embryo staining experiments show that the simulated microgravity effect can induce the differentiation and migration of neutrophils in zebrafish embryos,induce more neutrophils in the early stage of directed hematopoiesis,and enable the migration of neutrophils to the thymus.The process slows down.The simulated microgravity effect can also induce the differentiation of zebrafish embryonic blood cells and changes in the migration of hematopoietic sites,resulting in symptoms of aplastic anemia during the directed hematopoietic phase.The results of gene expression profiling chip analysis showed that there were 116down-regulated DEGs and 92 up-regulated DEGs in human peripheral blood lymphocytes under simulated microgravity effect.The results of GO functional enrichment analysis show that the cell components that have undergone significant enrichment mainly exist in the cell membrane,nucleus,cytoplasmic matrix and various organelles in the cell;the biological processes that have undergone significant enrichment mainly include intracellular biological reactions and intracellular Environmental homeostasis and the process of proliferation and differentiation;themolecular functions where significant enrichment occurs are concentrated in two parts:intracellular molecular binding function and intracellular protein activity regulation.The enrichment analysis results of KEGG signaling pathway indicate that the enriched signaling pathway is mainly related to the cell metabolic activity,growth and development process and the maintenance of homeostasis.Through the construction of the PPI network and the identification and analysis of gene modules,the selection of eight key genes that play an important role in the PPI network,CDKN3,NDC80,CDK1,NCAPG,BUB1,MAD2L1,CCNA2,and DLGAP5,was completed.Structural and functional analysis results indicate that these proteins are generally involved in mitotic cell cycle activities.
