According to the China tea gardens survey,the spiders of tea garden is one of the largest predatory natural enemy species in tea garden ecological system.Spider is belonging to arthropoda in animal taxonomy of arachnids spider.According to the World Spider Catalog Version 16(may,6,2015)reported that the spider is a total of 114 families,3,949 genera and 45,396 species around the world.Tea garden spider generate more than about 65%of tea plantations predatory natural enemy.Therefore,the study of spider communities and predation effect of tea pests in tea garden is very important.Especially,emerging molecular biology method is applied to evaluate the spider of tea garden controlling the main pests in tea plantations,it is huge scientific significance.For the tea plantation ecosystem in biological control to build the sustainable development and pollution-free green tea.First,this research study the condition of evarcha albaria predated tea looper(Ectropis oblique)and tea caterpillar(Euproctis pseudoconspersa)in different instars and the predation rate about Evarcha albaria predated tea looper and tea caterpillar under different starvation time in the field.The results showed that evarcha albaria has obvious selectivity for different ages,only prey on age 1 and 2 tea looper;With hunger time growing,the predation rate about tea looper is on the rise.When evarcha albaria hunger eight days,the spider prey on ages of age 1 or 2 tea looper,predation rate has reached 70.00%.But the spider do not feed on any age of tea caterpillar in this study.This research intuitive proves that the spider in the wild tea geometrid is feeding.The second,this experiment selects the Evarcha albaria as predators,Ectropis oblique as the spider 's prey to study spider 's feeding and digestion.we design a pair of specific primers According to a 657bp fragment of tea looper(Ectropis oblique)from the Gene bank.Using DNAMAN tool design a pair of specific primers Ectropis oblique F:5'-TGGAGGATTTGGAAACTGAT-3',Ectropis oblique R:5'-CCACCATGAGCAATAT TAGAG-3',The PCR product is 195bp fragment.The results proved that the primers are specific among 30 kinds of common tea insect pests,Euproctis pseudoconspersa and Empoasca vitis Gothe.This research designs a total of 16 time gradient that are 0.08 h,0.17 h,0.25 h,0.5 h,1h,2h,4h,8h,12h,24h,36h,48h,72h,168h,192h,360h.Each team has 10 samples repeat.Using PCR detection the rate of tea looper positive between spider cephalothorax,abdominal and foot segments and calculate the digestion half-life.The results shows that the spider digestion half-life is 284.44h.This research adopted two different sampling methods which include the trap method and clap nets method to investigate the spiders and Ectropis oblque in Wang Dazhen tea garden which located in hubei province.it was a period of 20 months,every 10 days for a continuous survey.Trap method collected total 8841 spiders,which belonging to 20 families,32 species,the dominant species is Zelotes asiaticus in trap method;Clap nets method collected 4194 spiders,belonging to 22 families,52 species,the dominant species is Evarcha albaria in clap nets method.Using correlation matrices to analyze the spiders which Collected by two methods and main pests.The results show that there is correlation between the spiders collected by trap method and Ectropis oblique,which indicated that Evarcha albaria has potential control function on tea looper in tea garden.
