| Spiders are important predators in tea-field ecosystem. The correct evaluation of the relationship between tea-field spiders and insects pests, the predation of spiders on pests have a very important significance for protection and utilization of the tea-field spider.The spider,belonging to Araneae (Arthropada Arachnida) is very common predators of insect pests in tea-field. According to Platnick (Platnick,2013) spider has a total of112families,3924genus and44540species around the world.Among them, Oxyope sertatus,Hylyphantes graminicola, Evarcha albaria, Misumenops ticuspidatus and so on has a great control effect on tea pests.Investigation results of tea-field show that tea-field spiders is the largest number of a group of tea garden of predatory natural enemy population groups, which generates more than about65%of predatory natural enemies.Therefore, we use molecular technology to study the spider’s feeding and digestion has very important scientific significance for bio-control of tea gardens.In this experiment, the author selects the Salticidae Evarcha albaria as predators, Empoasca vitis as the spider’s prey to study feeding and digestion, then, design a pair of specific primers According to a773bp fragment of Empoasca vitis that search from the Gene bank of NCBI, Gene bank:KC172507.1.Use gene tool design a pair of specific primers: Empoasca vitis F:5’-AGGTGCTGTATTTGCTAT-3’, Empoasca vitis R:5’-CTAAGAAATGTTGAGGGA-3’,The product is146bp.The results confirmed that the primer is specific among common18kinds of common tea insect pestsã€Euproctis pseudoconspersa and Ectropis oblique hypulina.Design0h,2h,4h,6h,8h,10h,12h,16h,20h,24h,36h,48h,72h,a total of13time gradient of10samples repeat gradient experiment of feeding time, distinguish between spider cephalothorax and abdominal segments of testing purpose fragment respectively in the positive rate and calculate the half-life.The results showed that at0h,10samples of repeated cephalothorax test positive rate was100%, the abdomen was0,2h after feeding the all fragments from chest liquid to the stomach and72h after testing positive for free sample, the results shows the purpose fragment in the spider has a half-life is42.92h.In the research, the author selected Wang Dazhen tea garden as survey sites which located in hubei province, we adopted two different sampling methods which include the trap method and clap nets method to investigate the spiders and Empoasca vitis, it was a period of20months,every10days for a continuous survey. Trap method collected8841spiders, which belonging to20families,32species, which the dominant species is Zelotes asiaticus; Clap nets method collected4194spiders, belonging to22families,52species, which the dominant species is Evarcha albaria. The author use correlation matrices to analyze the spiders which Collected by two methods and main pests, the results show that there is correlation between the spiders Collected by trap method and Empoasca vitis, which indicate that the spiders in tea garden have potential control function on the occurrence of the Empoasca vitis. |
