It's important to investigate and analysis the resources of Cordyceps sinensis and related fungi. r DNA ITS sequence analysis technology is useful to identify the phylogenetic status of Cordyceps fungi and other species of the genus accurately and quickly. According to the research of Yao Yi-Jian, Cordyceps' s pseudogene sequences impacted the accuracy of the system and construct the phylogenetic tree interference, which will generate different length and mistake phylogenetic tree constructed. It can leads to inaccurate results. ITS sequence analysis, morphological analysis and molecular phylogenetic analysis of the EF-1gene loci were carried out in the research center of Anhui Agriculture University.1. The experiment of several Ophiocordyceps sinensis fungus ribosomal gene transcription spacer(internal transcribed spacer ITS) on NCBI for sequence alignment and analysis. Metarhizikum taii, Paecilomyces gunnii, Codeceps militaris and Ophiocordyceps sinensis from Anhui agricultural university fungus library were determined ITS sequence and compared sequence characteristics to analysis the sequence features, based on r DNA technology. The system of the evolutionary tree was built, according to ITS characteristic sequences in Gen Bank database- nucleic acid homology ratio on retrieval. The results showed that cordyceps militaris Codeceps militaris and Paecilomyces gunnii have almost the same record information in strains library, similar rate is 98% to 100%.The results of Hirsutella sinensis showed that similar rate is 98%- 100%, too. But in the phylogenetic tree of Hirsutella sinensis,long and short branch was separated and penetrated between species, illustrated ITS sequence of pseudogenes. From the results, Metarhizikum taii, Paecilomyces gunnii,Codeceps militaris and Hirsutella sinensis are consistent in ITS analysis.2. Ophiocordyceps formosana compared with Beauveria bassiana sequences in Gen Bank database- comparing ITS, similar to the rate of 100%.After the morphology observation, results according with Ophiocordyceps formosana. Through analysis alpha loci in the E 1 molecular phylogenetic, the results is at 99% similar to Ophiocordyceps. According to the morphology and EF- 1 alpha gene loci analysis results can be obtained that is Ophiocordyceps formosana, without the possibility of the experimental sample contamination. It is concluded that Ophiocordyceps formosana on ITS analysis produced inconsistencies.
