Light is the most important ecological regulatory factor for shade plants,not only affecting the photosynthesis and material production,more importantly the regulation of many physiological and biochemical metabolic processes of plants,inducing and stimulating the activity and expression of key genes.Five colors of filter membrane were selected to study the effects of different light quality on the Baphicacanthus cusia,the results are as follows:1.Effects of different light quality on the growth morphology of Baphicacanthus cusiaRed film(R)is conducive to the growth of the aerial parts of the plant,18.08%higher than the control(CK).Transparent film(W)is conducive to the elongation of the root,,as well as the increase in fresh weight of plant.Chlorophyll a,chlorophyll b and chlorophyll a+b content are maximum under blue film(B)treatment,66.94%,90.70%and 76.07%higher than the control(CK)respectively.The net photosynthetic rate is the highest under transparent film(W)treatment,1.12 times of the control(CK).The trend of transpiration rate and stomata guide is consisten,the data are the largest under blue film(B)treatment both of two parameters.Yellow film(Y)is more conducive to the formation of soluble protein.The composite light quality is more conducive to the formation of soluble sugar.The protective enzymes activity in leaves is higher under red film(R)and blue film(B)treatment.2.Effects of different light quality on the active ingredient content of Baphicacanthus cusiaThe indigo content in leaves of Baphicacanthus cusia is higher under yellow film(Y)treatment,respectively,1.08 times,1.24 times and 1.28 times of the control(CK)in the 3d,7d and 15d.The indirubin content is the highest under yellow film(Y)treatment in the 7d,is 14.70%higher than the control(CK).The adenosine content is the highest under red film(R)in the 36d,2.80 times of the control(CK),so the long-term coverage of red film(R)may be conducive to the formation of adenosine.Overall,yellow film(Y)is more conducive to the formation of indole glucoside in 3d and 23d,55.97%higher than the control(CK)in the 23d.3.Effects of different light quality on the indole synthesis metabolic pathway related enzymes of Baphicacanthus cusiaThe TSB activity in leaves is highest under blue film(B)treatment,respectively,1.05 times,1.10 times,1.34 times and 1.49 times of the control(CK)in the 3d,15d,23d and 36d.The TDC activity in leaves is higher under yellow film(Y)treatment in the 3d and 7d,highest under red film(R)after the 15d.The AS activity in leaves is highest under red film(R)treatment,respectively,1.08 times and 1.01 times of the control(CK)in the 1d and 3d,the AS activity under composite light quality is higher in the 7d,15d and 36d.The ?-glucosidase activity in leaves is highest under purple membrane(P)treatment,1.71 times of the control(CK)in the 3d,the ?-glucosidase activity under composite light quality is higher in the 15d,23d and 36d.4.Effects of different light quality on the indole synthesis metabolic pathway related enzymes gene expression of Baphicacanthus cusiaThe TSB gene expression in leaves is highest under blue film(B)treatment,respectively,4.17 times and 1.87 times of the control(CK)in the 15d and 23d.The TDC gene is induced upregulation expression by purple membrane(P),respectively,3.18 times,1.58 times and 12.07 times of the control(CK)in the 1d,15d and 36d;red membrane(R)may inhibit the expression of TDC gene.The AS gene is induced upregulation expression by transparent film(W)in the 3d,7d,23d and 36d,yellow film(Y)makes AS gene expression down.The ?-glucosidase gene expression in leaves is highest under blue film(B)treatment,respectively,2.41 times,2.42 times and 15.26 times of the control(CK)in the 1d,3d and 36d.5.The correlation analysis of genes relative expression with active ingredient contentUnder transparent film(W)treatment,TSB gene relative expression shows highly significant positive correlation(P<0.01)between indirubin content,TDC gene relative expression show a significant negative correlation(P<0.05)with indigo content,?-glucosidase gene relative expression has a significant positive correlation(P<0.05)with indirubin content,AS gene relative expression shows a significant negative correlation(P<0.05)between indole glucosinolate content.Under blue membrane(B)treatment,a significant negative correlation(P<0.05)shows between AS gene relative expression and indole glucosinolate content.Under purple film(P)treatment,there is a significant negative correlation(P<0.05)between ?-glucosidase gene relative expression and indole glucosinolate content,both the relative expression of TDC gene and AS gene have a significant negative correlation(P<0.05)with the content of indirubin.Under red film(R)treatment,both the relative expression of TSB gene and?-glucosidase gene have a significant positive correlation(P<0.05)with the content of indirubin.Under yellow film(Y)treatment,there is a significantly negative correlation between AS gene relative expression and the content of indirubin(P<0.05).
