The research includes two parts:one is the solid-state fermentation condition of Penicillium xfsf-8 and the resistance to Ralstonia solancearum,the other is the control efficiency of solid-state fermentation inoculants of Penicillium xfsf-8 to Ralstonia solancearum and the control efficiency of liquid fermentation medium of Penicillium xfsf-8 to tobacco virus disease(TMV)in field.Tobacco mosaic and bacterial wilt is the main disease of tobacco.The incidence of virus disease in the nega is commonly 5%-20%,the person that weigh up to 50%,late onset the tobacco plant grow slowly and severe stunting.The tobacco is caused by Ralstonia solancearum,typical symptom is leaf blight,particularly in tobacco seedling stage often leads to tobacco seedlings wither and cause plant death.Penicillium xfsf-8 is separate from soil of tobacco field,flat resistance experiment showed high resistance to Ralstonia solancearum.Through single factor experiment,Plackett-Burman experiment,the steepest climbing and Box-Behnken experiment,the Penicillium xfsf-8 spore production condition of solid-state fermentation is optimized.according to the results of single factor experiment,in screening culture medium,corn flour as the carbon source,(NH4)2SO4 as the nitrogen source,yeast powder for grow factor,MgSO4 for inorganic salt,quantity of 1.2 x 107/ml,moisture content 40%,pH4,cultivating cycle for 8 days,1%twain-80 content in each group can achieve the maximum number of spores.By Plackett-Burman experiment select the significant influencing factors to Penicillium xfsf-8 solid-state fermentation spore production quantity,the results showed that yeast powder,water content,twain-80 as the significant influencing factors.Once Significant influence factors is determined,through the steepest upHill experiments determine the concentration of the yeast powder,twain-80,and water content,the results showed that when the 0.6%of yeast powder,3.28 ml of water content,0.9%of twain-80,the spore production can achieve the highest.With Penicillium xfsf-8 solid-state fermentation amount of spore production as the main target,based on the steepest experiment to determine the response surface experiment center and procee Box-behnken experiment,the results showed that when the 0.61%of yeast powder,3.77 ml of water content 0.8%of twain-80,the spore production can achieve the highest.Finally we get the best level combination of various factors:0.5%com flour as carbon source,0.5%(NH4)2SO4 as the nitrogen source,0.5%MgSO4 as inorganic salt,0.61%yeast powder as growth factor,quantity of 1.2 x 107/ml,moisture content 37.7%,pH4,0.8%twain-80 content and cultivating cycle for 8 days.By conducting different reagents on tobacco bacterial wilt in vega,the results showed that Penicillium xfsf-8 inoculants have a significant effect on tobacco bacterial wilt,the control effect was 87.15%,significantly reduced the incidence and disease index of tobacco bacterial wilt,the effect is better than Streptomycin sulpHate.By conducting different reagents on Tobacco virus disease in tobacco field,the results showed that Penicillium xfsf-8 inoculants have a significant effect on tobacco virus disease,the control effect was 60%,significant reduced the incidence and disease index of tobacco virus disease,the effect is better than morpHoline hydrochloride gung.
