Solid-state fermentation is effective and reliable to settle the resources shortage of Cordyceps militaris and the pressure from increasing market demand. Rice, a limited resource, is now used as substrate for large-scale fermentation. Soybean is used as an alternative for cultivation to multiply fermentation substrates. In order to lessen production time and yield new metabolites, the Cordyceps militaris fermentation system is optimized.The achievements arc as follows:1.Liquid fermentation was employed to choose Polysccharides high-yielding Cordyceps militaris strain:CM5.2. The optimum conditions with soybeans as medium for CM5 are:sample loading quantity 70g, nutrients 9ml and pH 6.0.3.HPLC was used to determine the ATP contents of Cordyceps militaris from conventioanl liquid fermentation, solid-state fermentation by rice and soybean. The results of ATP were 0.2546%,0.042% and 0.3062%,respectively.4.Orthogonal experiments were used to optimize the Polysccharides extraction procedure. The result showed that the extraction temperature, time, the ratio of raw material to water and time were 80℃,2h,1:20 and 3.The Polysacharides content was 6.93%.5. DPPH clearance test was utilized to sutdy the activity of Polysaccharides. The result showed that the clearance of undisposed Polysaccharides is higher preconditioning Polysaccharides. The Polysaccharides idicated high anti-oxidative activity.6.In order to study the Polysaccharide composition, Polysaccharides were seperated and purified.Three glycoprotein peaks 3were detected and named as follows:CM-Ⅰ,CM-Ⅱ,CM-Ⅲ. CM-Ⅱis subjected to derivatization test.
