As living standards improves,people pay more attention to the living condition and health problem,so more need to raise the requirements for the living environment and food safety.Currently,in order to prevent the disease of livestock,the abuse of antibiotics has become a common event in its breeding.Antibiotics played a major role in the development of animal husbandry and disease prevention.However,the potential damage of antibiotic on humans body health is more prominent.In china,the widely usage of antibiotic on livestock breeding leaded to less competition in the market of livestock and its products.Safe,environmental friendly and healthy forage additives,the alternatives of antibiotics,with the ability of disease prevention,had a promising development.The microecologics were made from probiotics which were screened out in the rumen of ruminants.The widely usage of microecologics on livestock breeding not only promoted the healthy growth,but also ensured food safety and humans body health.In this study,the probiotics was separated and purified in the fresh contents of bovine rumen from the slaughterhouse.The screened strains were studied on the characteristics of producing enzyme.The strains with higher enzyme production were identified to carry out the research about physiology and biochemistry analysis and 16 S rDNA.The experimental results are as follows:Firstly,the bacterial suspension after enrichment culture was diluted with a gradient,separating and purifying 28 kinds of strains,including 19 kinds of bacteria,numbered X1 to X19 and 9 species of fungi,numbered Z1 to Z9.Secondly,the separated 28 strains were conducted on preliminary screening of enzyme production strains by differential medium,screening out cellulase producing strains: X1,X2,X3,X4,X8,X9,X11,X12,Z7,Z8,Z9,a total of 11 species and amylase producing strains: X1,X3,X4,X9,X10,X11,X12,X13,X14,X15,X16,X17,X18,X19,a total of 14 species.Thirdly,by comparing,the strains that produce two enzymes with higher enzyme activities were X3 and X4 strains.These two strains were carried out the measurement of enzyme activity respectively,and the results were that cellulase activity of X3 was 60.38 U/mL and amylase activity was 53.94 U/mL,the cellulase activity of X4 was 43.22 U/mL and amylase activity was 46.19 U/mL.Fourthly,X3 and X4 strains were carried out physiological and biochemical identification and 16 S rDNA strain identification respectively,identified that X3 strain was starch solution bacillus,and X4 strain was bacillus subtilis.According to the regulations issued in the “catalog of feed additives varieties” in 2013,the starch solution bacillus could be used as related enzymes in feed and bacillus subtilis can directly be added into the feed as microbial additives.So the strains X3 and X4 by screening in this study are beneficial to rumen microbes,which play a positive role in the development of microecological of ruminants.
