Autolysis phenomenon exists in all kinds of microorganisms widely.In this paper,different carbon sources culture were used to induce the autolysis of Pleurotus tuber-regium Differential proteomics and metabonomics were conducted to study the autolysis mechanism.1.Different carbon sources culture were utilized to induce the autolysis of Pleurotus tuber-regium and the optimization of its relevant condition were carried out.It was suggested that the strong autolysis phenomenon of Pleurotus tuber-regium happened in the follow conditions,i.e.training mode of first glucose then corn starch,buffer system of pH 7,keeping warm at 40 ?.2.Comparing the extraction method of Pleurotus tuber-regium's hypha protein,it showed that the protein samples extracted by phenol method were optimal to conduct dimensional electrophoresis.Consequently,phenol method was applied to extract the Pleurotus tuber-regium protein in different carbon source culture for the differential proteomics research.3.The results of differential proteomics showed that there were 6 proteins related to energy metabolism,9 proteins relevant to nucleic acid metabolism,8 proteins related to protein metabolism,15 proteins related to material transportation and signal transduction,6 proteins relevant to material metabolism appeared differential expression.It was demonstrated that the autolysis of Pleurotus tuber-regium was influenced by the carbohydrates synthesis.In the first glucose then corn starch cultivating mode,Pleurotus tuber-regium was restricted by temperature and available monosaccharide so that its autolysis was severe relatively and multiple signal transduction pathways enhanced.Biological macromolecular synthesis was promoted largely,in turn contributed to improving the Pleurotus tuber-regium's tolerance to the environment.In first corn starch then glucose training mode,the carbon utilization of Pleurotus tuber-regium shifted from corn starch to glucose so that its autolysis was gentle and most of the biological membrane functions remained.Whereas in glucose as sole carbon cultivating mode,the original carbon utilization mode of Pleurotus tuber-regium had no alternation,its autolysis was influenced almost only by temperature so that the heat shock proteins were in the majority in the signal transduction pathway.4.Metabonomics were conducted to study the autolysis of Pleurotus tuber-regium in different carbon source cultivating modes.22 materials were identified,in which both monosaccharide varieties and relative quantity went up.In conclusion,it was inferred that the autolysis mechanisms of Pleurotus tuber-regium were as follows.In deficient available monosaccharide,the normal metabolism balance was broken.Then in response to the external environment change,glycosidase and protease were synthesized,part of the signal pathway expressed,all of which work together to degrade the cell wall polysaccharides and part of the cytoplasm components so that the microorganisms began to autolysis.The nutrition material obtained from autolysis was applied to Glycolysis pathway to synthesize biological macromolecular when ATP was supplied and in turn assembled into new cell to repair the original cell.Pleurotus tuber-regium grew normally when the synthesis reaction was faster than autolysis reaction.
