T.gondii and Chlamydia,as two kinds of serious zoonotic pathogens,pose a great threat to the health of human beings and survival of animals.The study of two kinds of pathogens in the wild animals and pet birds includes infection,analysis of variation rule of interspecific relationship and toxicity of T.gondii,which has a guiding significance for prevention and treatment of Toxoplasma gondii.1.Investigation and Analysis of Serum antibody of Toxoplasma gondii and Chlamydia for bred wild animals in Fuzhou zoo.The modified agglutination test(MAT)was carried out to detect the antibody level(IgG)of T.gondii in 27 species of 132 mamals and birds in the Fuzhou Zoo and 123 rhesus macaques in the Family Planning Institute.The results of investigation discovered that the positive rates of antibodies to T.gondii of wildlife in the Fuzhou Zoo and 123 rhesus macaques were 23.48%(31/132)and 4.88%(6/123)respectively.The wild animals of T.gondii were treated with Sodium sulfamonomethoxine,Folic acid and delta-Gluconolactone.The result of drug testing showed that the effective rate of the 9 positive animals of T.gondii was 77.78%;and the negative conversion rate was 33.33%,showing the obvious effect of the drug experiments.The calculation of the original data of antibody level of zoo animals,the variance value is larger than the average value,belonging to aggregated distribution,it showed that the improve risk of infection rate increase in the Fuzhou Zoo.The serum antibody level(IgG)of Chlamydia in 27 species of 132 animals in the Fuzhou Zoo and 123 rhesus macaques in the Family Planning Institute were detected by the indirect hemagglutination test(IHA).The results of investigation showed that the positive rates of antibodies to Chlamydia of wildlife in the Fuzhou zoo and rhesus macaques were 24.42%(32/132)and 3.25%(4/123).The antibody level of Chlamydia of zoo animals is low,less than 1:64,and the uniform distribution and the variance value are less than the average value,belonging to evenly distributed,and it showed that Chlamydia infection is relatively stable.The Reciprocal and Logarithm of antibody titers of T.gondii and Chlamydia were analyzed by correlation and regression analysis,and the result discovered that there was significant negative correlation between the T.gondii and Chlamydia(r=0.6901,P<0.01);and the correlation analysis also showed that the negative association between Chlamydia and T.gondii(V=-0.104887),showing that presenting a certain degree of mutually exclusive relationship existing between Chlamydia and T.gondii in zoo animals.2.Genetic Characterization of Toxoplasma gondii from Bred Wildlife in FujianHeart tissues were collected from 204 animals from zoo,pet market and other place.After identified by using B1 gene,the positive samples of T.gondii were typed at 13 genetic markers,including SAG1,5,and 3'-SAG2,SAG2,SAG3,BTUB,GRA6,c22-8,c29-2,L358,PK1,Apico,and CS3.The results showed that 13 samples were found to be positive by polymerase chain reaction(PCR)amplification using T.gondii B1 gene primers,including 7 zoo animals(1 Black-capped,1 Wild Red Dog,1 Lemur,1 zebra,1 Red panda,1 Peacock,1 Red-crowned Crane,15.56%,7/45),3 pet birds(1 Budgerigar,1 Black-billed Magpie,1 Zebra Finch,2.14%,3/140)from pet market and 3 cats(15.79%,3/19).Due to positive samples,the results were classified into four distinct genotypes:ToxoDB#9(Black-capped),ToxoDB#2(Peacock)and ToxoDB#10(Budgerigar)and New Genotype(cat),respectively.This is the first genotyping of T.gondii from zoo wild animals and pet birds in Fuzhou.MEGA5.0 software to carry on the income of phylogenetic tree was used for phylogenetic tree analysis of amplified gene sequences of SAG1,SAG2 and SAG3 of T.gondii were amplified and GenBank database,the results were the same with the PCR-RFLP results.The modified agglutination test(MAT)and The indirect hemagglutination test(IHA)were carried out to detect serum antibody level to T.gondii and Chlamydia of 132 animals from zoo and 123 rhesus macaques in the Family Planning Institute respectively,with the drug experiment for the part of T.gondii positive animal and using the method of PCR-RFLP to genotype T.gondii from positive animals in zoo animals,pet birds and cats in Fuzhou.The study has certain guiding significance for prevention and treatment of T.gondii in zoo animals,pet birds and cats,and also has an important significance in public health.
