| Tripterygium wilfordii(Tripterygium wilfordii Hook.F.),a kind of precious medicinal plants,is mainly distribut in China’s Fujian,Zhejiang,Anhui,Jiangxi,Taiwan and other provinces.Endophytic fungi were widely exists in plant,form a mutually beneficial association in the long-term living with the host plants affect each other.In the micro ecosystem,they live for a long time with their host plant.Related studies have shown that,Endophytic fungi can promote plant growth,enhance the host plant resistance,strengthen the metabolism of the host plant and the many kinds ofbiological function,someendophytic fungican compound secondary metabolite which same or similar host plants.Recently the source of tripterygium wilfordii are mainly composed of excavation the wildlife resources,but tripterygiu wilfordii resource is limited,this phenomenon is result that ripterygium wilfordii raw resources reducing and effect on the local biodiversity and ecological balance.According to build co-culture ecosystem,Evaluated the endophytic fungi beneficial biological activity and technology of suspended inducer.Research effect of endophytic fungi isolated from Triperygium wilfordii on the growth and active ingredients biosynthesis of host plant.Main results are as follows:1.Put the 18 endophytic fungi in the vaccine,there are 7 strains had inhibiting effect of vaccine.NS1,NS6,NS31,NS32,NS33trains not only promote the growth of Tripterygium wilfordii,but also could improve the triptolide contents and tripterine of different organs significantly.In physiological and biochemical aspects,NS6,NS7,NS 14,NS32 strains had high activity of produce phytohormone;NS-1,NS-31strains were capable of dissolving phosphorus.NS1,NS4,NS6,NS25,NS31,NS33strians had synthetic iron carrier ability.Comprehensive indicators choose NS6,NS31,NS32,NS33 strains as target strains.2.Bacterial 26S rDNA sequence identification and homology analysis suggested that NS6 belonged to the penicillium steckiian;NS-33strains belonged to the hypocrea nigricans;NS31,NS33 belonged to the meyerozma gulliermondii and fusarium oxysporum.All their similarity was 99%.3.Through made 4 strains to co-culture and put they in the vaccine,they only could promote the growth of Tripterygium wilfordii but also improve the C、N、P、K element contents of different organs significantly.Improve chlorophyll contents and Fv/m value.All kinds of mix strains could promote the triptolide and tripterine contents.Effect on The NS31-33 strains the triptolide content is the highest and Effect on The NS6-32、NS6-33 strains the tripterine contents were higher than others.4.Make 4 target strains into bacterium liquid and added to tripterygium wilfordii suspension cells.The results show that:all elicitor could improve the consumption rate of nutrients suspension cell;except NS33-JT and NS31-JY inducers others inducers can improve suspended cell vitality;and in the NS32-JT、NS33-JT inducers influence,cells weigh were lower than other treatment;Intracellular triptolide content in NS31-JY and NS32-JT were higher than others;in NS32-JT and NS31-JT inducers influence the rate of synthesis ttripterine had improved;in the culture did not found the triptolide.5.Evaluate the endophytic fungi beneficial biological activity,choose NS6-31 mix strains make into 3 kinds of inducer:endophytic fungi bacterium liquid(JY),crude extractings from endophytic bacteria(JT)and untreated living bacterium(HJ).We added these three kinds of inducer into tripterygium wilfordii suspension cells,and the results showed that the three kinds of inducer can increase the soluble nutrition consumption rate in suspension cell cultures;HJ inducer can increase the culture solution PH value but JY and JT could make it gone down;all kind of inducers were reduced the nutrient solution electrical conductivity;JY and JT inducers could improve suspension cells weigh,HJ were contrary;JY inducer could improve the triptolide content;JT and JY inducers could improve tripterine content,the result is the same with chapter 4 that in the culture medium did not found tripterine. |
