Bucephalandra sp."Kudangan" is a kind of tropical aquatic plants of Araceae Bucephalandra.In recent years,they have been attracted widespread attention as characters in shapes and colors of their leaves are different when terrestrial and aquatic,and only in water,the colors of their leaves are flexible.In this experiment,for the purpose of establishing rapid propagation of Bucephalandra sp."Kudangan",we used its stem apex,stem segments with axillary buds as the research materials.The results show that:(1)When used shoot tips as explants,after sterilization with 75%ethanol 30s,1.5%NaClO 2min+0.1%HgCl22min treatment was the optimal sterilizing type,MS+6-BA 1 mg/L+NAA 0.3 mg/L of adventitious bud induction medium was better than others;(2)When used stem with axillary buds as explants,after sterilization with 75%ethanol 30s,1.5%NaClO 4min+0.1%HgC12 2min treatment was the best sterilization method,MS+6-BA 1 mg/L + NAA 0.5 mg/L for adventitious buds induction;(3)Two types of explants material used the the subculture medium with the most suitable medium in(1)and(2);(4)1/2MS culture medium as root induction.The above all cultured in 25?,1500-25001x illumination 12h/day can produce complete plants.The rapid propagation of plants were obtained before transferred to hydroponics,here we show the observation of leaves color change in different light conditions and CO2 concentrations:1.High light intensity and long illumination time can obviously promote the anthocyanin production,and increased content of chlorophyll a,b,also promote the increase ratio of chlorophyll a faster than chlorophyll b.So the plant leaves appeared manifest by blue green color,leaf surface layer of brown metal color gloss.2.Different concentrations of CO2 on anthocyanin and chlorophyll content is very obvious in the beginning,but will no longer continue to go up.In addition,we find in the same aquarium conditions,leaf growth increased significantly with CO2,so the effect to the colors of leaves is not obvious to Bucephalandra sp."Kudangan".
