| From Tai’an City,Shandong Province,well-grown tea Cherry Orchard collected rhizosphere soil,the use of selective media for solubilizing bacteria,silicate bacteria and nitrogen-fixing bacteria were isolated screening;to Rhizoctonia solani and Fusarium Fusarium as the target pathogen screening antagonistic bacteria isolated from tea rhizosphere bacteria;biological activity of various growth-promoting bacteria were measured and studied the biological activity of the higher bacterial isolates in the rhizosphere colonization tea case of bacteria can colonize isolates were identified.The main results are as follows:(1)through six tea root soil samples collected for processing the occasion of solubilizing bacteria isolated 67 isolates,71 bacterial isolates silicate,63 nitrogen-fixing bacteria isolates,66 isolates antagonistic bacteria.(2)Biological activity of each growth-promoting bacteria test showed that,4T-P-5,4T-P-10,5T-P-10,5T-P-14 isolates phosphate solubilization capacity stronger after 72h culture,media the increased amount of soluble phosphorus between 93.5ug/mL-244.3ug/mL,achieve functional microbial fertilizer production strains USDA requirements;TK-7,4T-K-4,4T-K-21 isolates high potassium solution capabilities,medium K+content increased by 43.1%,respectively,53.4%and 44.42,to the functional requirements of microbial fertilizer production strains prescribed by the Ministry of Agriculture;4T-N-11,4T-N-12,5T-N-8 high performance nitrogenase isolates,1g glucose consumption of fixed nitrogen were 22.2ug,27.8ug,35.5ug;T-10,3T-4,4T-22 isolates against Rhizoctonia solani and Fusarium oxysporum the antagonistic effect is good.(3)the use of antibiotic marker methods to study the characteristics of high biological activity colonization of 13 isolates in the rhizosphere of tea,a total of 12 isolates were able to colonize the rhizosphere of tea.(4)to be able to isolate the tea rhizosphere colonization of mutual antagonism test results show that,5T-P-10 and 4T-P-10 isolates antagonistic phenomenon,4T-N-11 and 5T-P-10 isolates have antagonistic phenomenon,4T-K-21 and 4T-N-12 isolates antagonistic phenomenon,and the rest did not antagonistic bacterial isolates phenomenon.(5)not mutually antagonistic strains isolated phenomenon hemolysis test results show 4T-P-5,5T-P-14,3T-4,5T-N-8,TK-7,4T-K-4 no on blood agar plates transparent circle,other isolates with a transparent circle,determine the initial bacterial isolates these six is safe available.(6)integrated morphological,physiological and biochemical characteristics and 16S rDNA sequence analysis of six growth-promoting bacteria in the rhizosphere of tea were identified,including solubilizing bacterial isolates 4T-P-5 as Paenibacillus,solubilizing bacterial isolates 5T-P-14 as Pseudomonas putida,silicate bacterial isolates 4T-K-4 as Serratia marcescens,silicate bacterial isolates TK-7 for the huge Bacillus,nitrogen-fixing bacteria isolates TN-8 Bacillus,antagonistic bacterial isolates 3T-4 class Bacillus. |
