The powdery mildew disease caused by Blumeria graminis DC f.sp.tritici EO Speer(Bgt)inflicts severe economic losses in wheat crops.It seriously affect the yield and quality of wheat.Therefore,it is very important to find the powdery mildew resistance genes.Receptor-like kinases(RLKs),a family of ubiquitous membrane proteins in higher plants,play important roles in plant growth and development and their responses to environmental stimuli,The RLK10 receptor like protein kinase is a typical type of receptor like protein kinase,according to the different types of structure domain of plant receptor like protein kinase.It was first found in wheat,in the monocots are found to exist in the form of multiple gene copies.Up to now,there are few studies on the specific RLK10 family in Monocots.In this thesis,we used the RLK10 transgenic wheat to study the potential resistance mechanism of RLK10 to wheat powdery mildew.Here,two gene members TdRLK1 and TdRLK3 were used.RLK First,we used TdRLK1 transgenic tetraploid wheat to study the powdery mildew resistance of RLK10 gene family.It includes two aspects:1)Identification the transgenic lines to screened contain NP::TdRLK1(gDNA)-GFP and NP::TdRLK1(cDNA)-GFP transgenic tetraploid wheat.2)Based on the Molecular-detected disease index(MDX),we found that the transgenic wheat with NP::TdRLK1(gDNA)-GFP had stronger powdery mildew resistance than NP::TdRLK1(cDNA)-GFP transgenic wheat.We deduced that the TdRLK intron sequences may play an important role in the regulation of gene transcription.By using F2 population,we analyzed the powdery mildew resistance of TdRLK1and TdRLK3 transgenic materials from two aspects:1)Molecular identification the transgenic lines of tetraploid wheat.2)By using the in vitro inoculation with powdery mildew and fluorescence microscopy to observe the development of powdery mildew mycelium we selected two groups,W126*Pg12 F2 and W126*Pg19 F2,which were transgene with TdRLK1 and TdRLK3,respectively.Based on the difference of the MDX value,we found that TdRLK1 transgenic wheat had stronger powdery mildew resistance than TdRLK3 transgenic wheat.We speculated that TdRLK10 receptor like protein kinase involved in the resistance of wheat to powdery mildew.
